The Role Of NNOS In The Regulation Of Electro- Acupuncture(EA) Anti-biphasic Depression

Objective:The preliminary research progress on the influence of nNOS on the regulation of Electro-acupuncture(EA) anti-double state-anxiety and depression of depression.Methods:Experiment research 1:84 healthy male SD rats were randomly divided into 6 groups,control (1/2) group,Anxiety group,Depression group,EA anti-anxiety group(EAAA) and EA anti-depression group(EAAD).Anxiety and Depression group was produced by chronic unpredictable mild stress combined with separation for 7 and 21 days respectively, EA (100Hz and 2 Hz,1 mA) were performed at acupoint "Baihui" (DU20) and "Yintang" (GV29) for 15 min,once a day for 7 and 21 days, the control(1/2) group did not get any intervention.Experiment research 2:24 healthy male SD rats were randomly divided into 3 groups: Control,Footshock(FS),EA anti-anxiety(EA-FS) was produced by inevitability footshock Com-bined with separation, EA-FS EA (100Hz,1 mA) was performed at acupoint "Baihui" (DU20) and "Yintang" (GV29) for 15 min,once a day for 7days.Elevated plus maze(EPM) was carried out to evaluated Anxiety-like behavior and the open-field test,absolute relative volume of sucrose-intake and body weight were used to evaluated Depression-like behavior, reverse transcriptase polymerase chain reaction(RT-PCR), Westren-Blot(WB) and immune-histochemistry were used to detect the protein of nNOS expressed in hippocampal and CA3 and CA1 areas respectively.Results:Experiment research 1:1.The results showed that CUMS for 7 days signinificantly decreased the percentage of time and entries (P<0.01) in open arm compared to control 1 group in the EPM, EAAA can significantly increased them (P<0.01); CUMS for 21 days significantly decreased the hole horizontal movement distance and percentage of entries (P<0.01) in central area compared to Control 2 group in the Open-field test, EAAD can significantly increased them (P<0.01); On the 7th, the body weight of Depression group and EAAD were lower than that of Control2 group, but no significant difference was found between them, from the 14th to 21st day, the body weight of Depression groups were significantly lower than that of control2 group (P<0. 01), EAAD can significantly increased them (P<0.05);On the 7th, the absolute and relative volume of sucrose-intake of Depression group was lower than that of Control2 group, but no significant difference was found between them,from the 14th to 21st day, the absolute and relative volume of sucrose-intake of Depression groups were significantly lower than that of control2 group (P<0.01), EAAD can significantly increased them (P<0.05).2. PT-PCR:The nNOS mRNA expression in hippocampus in Anxiety and Depression group were lower then then that in Control(1/2) group, EAAA and EAAD can increased them;3. Western-Blot:The nNOS mRNA expression in hippocampus in Anxiety and Depression group was lower then that in Control(1/2) group, EAAA and EAAD can increased them;4.Immunohistochemistry:CUMS for 7 and 21 days significantly decreased the Density mean and IOD of nNOS in hippocampus CA3 and CA1 (P<0.05), EAAA and EAAD can reversed them (P<0.05)Experiment research 2:1.The results showed that inevitability footshock for 7 days significantly decreased the percentage of time and entries in open arm compared to Control group in the EPM, EA-FS can significantly increased them (P<0.05);â‘ The nNOS mRNA expression in hippocampus in FS group was higher than that in control group, EA-FS decreased them;â‘¢Repeated inevitability footshock significantly increased the Density mean of nNOS in hippocampus CA3(P<0.05), EA-FS can reversed them(P<0.05), in hippocampus CA1 they were significantly decreased(P<0.01), EA-FS can reversed them(P<0.01).Conclusion:The depression in different stress time-in the early chronic stress(CUMS for 7 days) and late chronic stress (CUMS for 21 days) showed different statuses well as anxiety and depression; the expression of nNOS would show different state as the different factor and intensity in stress accordingly,EA can caused anxiolytic-like and antidepressant-like effects, which may attribute to the regulation of nNOS on hippocampus.
