The Effect Of Hyperin And Icariin On The Expression Of CYP17 And CYP19 In Rat Ovarian Granulosa Cells

Objective:To investigate the effect of different concentrations of Hyperin and Icariin (ICA) on the proliferation and the secretion of estrogen(E2), progesterone(P) of GCs. To explore the mechanism of Hyperin and Icariin (ICA) affecting the steroid hormone synthesis by detecting the expression of CYP17 and CYP19.Methods:The ovary granulosa cells of rats are cultured in vitro and treated with Hyperin and Icariin, the growth curve of granulosa cells at different incubation times are evaluated. The number of ovarian granulosa cells observed under the microscope were counted. The proliferative quantity of ovarian granulosa cells was measured by MTT cell proliferation assay.The concentration of E2 and P in cultured liquid was assayed by ELISA. The CYP17、 CYP19 mRNA expression was detected by Real-time quantitative PCR and the expression of relevant proteins and the coded genes was examined by Western Blot.Results:Hyperin and Icariin could obviously increase the number of ovarian granulosa cells, and increase the secretion of estradiol (E2) and progesterone (P). Real-time quantitative PCR was used to detect the CYP17、CYP19 mRNA expression, the result indicated that:compared with the control group, most of the Hyperin and Icariin concentrations could observably increase the relative value of CYP17、CYP19 mRNA expression. Hyperin could increase the relative value of CYP17、CYP19 mRNA expression achieving the maximum effects at 50ug/L, while Icariin was at lOug/L. Western Blot was used to detect the protein expression of CYP17 and CYP19. The protein expression of CYP17 and CYP19 was mostly enhanced at the concentration of 50ug/L Hyperin and 10ug/L Icariin.Conclusions:Hyperin and Icariin could increase the number of ovarian granulosa cells cultured in vitro,promote the secretion of E2 and P,improve the relative value of CYP17、CYP19 mRNA expression and enhance the protein expression of CYP17、CYP19.So we concluded that some concentration of Hyperin and Icariin can increase the number of ovarian granulosa cells cultured in vitro and promote the secretion of E2 and P through up-regulating the CYP17 and CYP19 gene expression to stimulate the synthesis of steroid hormones.
