Objective:To investigate the role of ORAI1 in the injury-induced rat carotid artery neointimal formation and the relationship between ORAI1 and NCX1.Methods:Rat left common carotid artery balloon injury model was established and lentiviruses particle was constructed successfully. The ORAI expression was dectected using real-time PCR and western blot, and PCNA and NCX1 expression were measured by western blot. Neointimal formation was analyzed by HE staining.Results:The neointimal was formatted at 7 and 14 d after injury. Compared with the sham group, the expression of PCNA and ORAI1 was upregulated at 7 and 14 d after injury (P<0.05). ORAI1 knockdown inhibited the injury-induced neointimal formation (P<0.05). Meanwhile, NCX1 expression was also decreased by ORAI1 knockdown.Conclusion:ORAI1 has a key role in the injury-induced neointimal formation, VSMCs proliferation and NCX1 expression, and may be a therapy target for arteriosclerosis.
