Objective:To observe BDNF and TrKB protein in hippocampus and learning and memory in rat model of autism.Methods:Ten adult Wistar of male and female rats were selected,each one of male and female was caught in a caged at 19:00 together. Female rats that were found the vaginal plug recorded as embryonic 0.5 day and were feeded in cage alone. The pregnant rats were randomly divided into model group and control group. The rats in E12.5 of model group were given intraperitoneal injection of VPA ip600mg/kg,control group were normal feeding.The childrens of the model group were recorded as VPA rats; The normal group childrens called control rats.According to the development behavioral tests and selecting autism rats. After Morris water maze, test the rats of ability of learning and memory.Selecting 36 VPA rats and divided them into "changqiang" group, "feixue" group and model group, each group has 12 rats; Normal Wistar rats offspring randomly selected 12 rats as control group, "changqiang" group take "changqiang" point, "feixue" group choose the right flank fixed points,Electro acupuncture continuous stimulation (continuous wave, the frequency of 2Hz, Strength is 2mA),20min needle,one courses 20 times."feixue"group operation are similar with "Changqiang" group.The control group and the model group were reared under the same conditions without any intervention. At the end of treatment to test the Morris water maze, Immediately take the hippocampus to observe the expression of BDNF and TrKB protein by Western blot.Results:1.Morris water maze test:Before intervention:Compared with the model group, The control group escape latency shorter, there was significant difference(P<0.05); Compared with the control group:"changqiang" group, "feixue" group escape latency was prolonged (P<0.05); The number of cross platform had no obvious difference among the four groups. After intervention:Compared with the model group,"changqiang" group and control group escape latency was shorten (P<0.05), Compared with the control group:"feixue" group escape latency was prolonged (P<0.05); Compared with the model group, The control group and "changqiang" group across the platform number increased(P<0.05); Before and after the intervention each group had significant difference(P<0.05).2.Western blot:Compared with the model group, Blank and "changqiang" groups have higher relative volume of BDNF and TrKB protein in the hippocampus, While "feixue" group is Close to the model group (P<0.05).Conclusion:Electroacupuncture "Changqiang" can enhance the expression of BDNF and TrKB protein in hippocampus of VPA autism rats and improve their ability of learning and memory.
