| According to the new reports, peroxide play important role in a number of human neurological disorders.H202, as ordinary peroxide, can cause neurotoxicity in a certain degree of concentration. However, the mechanism is not clear.In order to reveal the molecular mechanism, PC12 was cultured with various time in culture medium contained different concentration of H2O2. The related genes of nerve growth, apoptosis and autophagy have been detected at transcriptional and translational level using real time PCR, western blotting. ROS and cell cycle were detected with Flow Cytometry (FCM), and the length of synaptse were tested by immunofluorescence (IF).Here we showed that mitochondrial ROS was increased depending on the added dose of H2O2. And when the concentration was higher than 200umol/1, cell cycle was arrest in G2/M phase and the length of synapse was altered shorter than normal cultured cell.In addition, with increasing concentration, the expression of beta â…¢ tubulin, NCAM which are nerve growth rerelated genes incrementally decreased, with increasing concentration, the transcription of NeuroDl and GAP-43, repair related genes were increased, as the time went on NeuroDl and GAP-43 were decreased.Moreover, as the time went on, the growth of synaptse was inhibited accompanied by increased caspase-3, Bax, inhibited Beclinl and lc3B, suggesting that H2O2 may cause the damage of PC12 through apoptosis and autophagy which can provide novel therapeutic clues to traditional neurological diseases treatment. |
