Yindan xinnaotong soft capsule (YDXNTSC), a compound preparation used in traditional Chinese medicine, is composed of 8 herbs:Ginkgo biloba leaf (yinxingye, YXY), Salvia miltiorrhizae (danshen, DS1), Herba gynostemmatis (jiaogulan, JGL), Erigerontis herba (dengzhanxixin, DZXX), Allii sativi bulbus (dasuan, DS2), Notoginseng radixe rhizoma (sanqi, SQ), Crataegi fructus (shanzha, SZ), and Borneolum (tianranbingpian, TRBP). In China, YDXNT soft capsules have been used to treat cardio-cerebrovascular diseases for several decades. However, until now there have been few studies of Pharmacodynamic Material Foundation. In this work, fingerprint, multi-component quantitation and bio-activity were utilized to clarify the constituents, absorbed chemical constituents, and pharmacological action of YDXNTSC. It laid foundation for clinic application. The main research contents are presented as follows:1. The UPLC-MS fingerprint analysis of YDXNTSCThe UPLC-Q-TOF-MS method was used to identify the chemical constituents of YDXNT. Approximately 74 components were detected, of which 70, including 23 flavonoids,4 ginkgolides,15 phenolic compounds,18 diterpenoid tanshinones, and 10 ginsenosides, were tentatively identified. In addition,10 batches of YDXNT were tested and compared. A fingerprint analysis was developed to evaluate the stability of YDXNT samples2. The multi-component analysis of YDXNTSCThe UPLC method was established to simultaneously determine 3 active components included tutin, scutellarin, and salvianolic acid B in YDXNTSC. The 3 constituents of ten batches of samples were simultaneously determined as well. This method was proved to be a rapid, reliable, and conveninent technique.3. The analysis of YDXNTSC absorbed chemical constituentsThe UPLC-QQQ method was used for determining the absorbed YDXNTSC solution. Firstly, the absorded YDXNTSC solution was acquired by using the everted gut sacs methord, and then the solution was test by UPLC-QQQ. The results showed that 11 constituents all could be absorbed by everted-gut sacs. This may be a reliable foundation method in pharmacological research in vitro.4. The bio-activity study based on the combination of everted gut and vasorelaxation activityThe combinative method of everted gut sacs and exsomatize blood pipe ring was estabolished to explorer the activity of vasorelaxation. The result showed that the absorded YDXNTSC solution possess antagonism casued by KCl vasoconstriction. Indicating the absorded solution has vasorelaxation. In addition, the effect will be better with the dose increased. This method could reflect the real bioactivity of YDXNTSC.
