Objective: To investigate the effect of BuNao capsule on memory,antioxidation and the hyperphos-phorylation of tau protein in D-galactose and Aβ25-35 induced Alzhheimer’s disease(AD) rats model. To provide experimental basis for clinical treatment of AD with BuNao capsule. Methods: Totally 90 SD rats were randomly divided into control group,model group, Piracetam group, BuNao capsule high,middle and low groups.The model rats were estsblished by injection of Aβ25-35 into the lateral cerebral ventricle and D-galactose(ip.).Then orally given drugs for 8weeks.The learning ability was determined by Morris maze test. The method of HE staining was performed to observe the changes in neuronal morphology and modified Bielschowsky silver staining was performed to observe the morphological changes of the nerve fibers in rat brain.The activities of glutathione peroxidase(GSH-Px),superoxide dismutase(SOD) and content of Malondialdehyde(MDA) in brain were measured with Elisa. The Tau protein which includes total-tauã€p-tau(Ser396)ã€p-tau(Ser404)in the neuron were determined by Western blot analysis. Results: 1.Morris maze test shows that the learning ability of BuNao capsule groups were superior than AD model group( P < 0.01). 2.The HE staining shows that the morphology of treatment groups were superior than AD model group; 3.The Bielschowsky silver staining shows that the neuron entanglement of treatment groups were superior than AD model group. 4.The activities of GSH-Px and SOD were declined,and the MDA content was increased in AD group compared with control group(P < 0.05). The activities of GSH-Px and SOD were heightened and the MDA content in BuNao capsule group declined remarkablely compared with AD model group(P<0.05).5.The phosphorylation level of total tau protein and its two sites tau(ser396/ser404) was significantly lower than that in model group(P < 0.05). Conclusion: 1. The method of intraperitoneal injection of D- galactose combined with bilateral ventricle injectionof Aβ25-35 can successfully establish the rat model of Alzheimer’s disease(AD).2.BuNao capsule can improve the learning and memory ability of AD model rats,improve the pathological changes of brain tissue, and have a certain therapeutic effect on AD model rats.3. BuNao capsules can enhance rat brain antioxidant enzyme activities of SOD and GSH Px, decrease the content of MDA, reduce neuronal tau hyperphosphorylation. It may be one of the mechanisms of BuNao capsule in the treatment of AD model rats. |
