Objective: To observe the effects of the total flavones of Mappianthus iodoies on myocardial ischemic-reperfused injury in rats. Methods: The coronary artery of seventy-two male SD rats was ligated to make into the model of acute myocardial ischemic reperfusion; they were randomly and evenly divided into six groups, they were the sham-operation group, the ischemia-reperfusion model group, the positive control drug group, the total flavones of Mappianthus iodoies with low does(60 mg·kg-1) group, the total flavones of Mappianthus iodoies with middle does(120 mg·kg-1) group and the total flavones of Mappianthus iodoies with high does(240 mg·kg-1) group. HE stain was used to observe the change of myocardial pathomorphology of rats;the change of myocardial infarction size(MIS) in rats were detected by TTC stain; the contents of malondialdehyde(MAD), superoxide dismutase(SOD),tumor necrosis factor-α(TNF-α), interleukin-6(IL-6), creatine kinase(CK) and lactate dehydrogenase(LDH) in serum of the rats were detected by ELISA; The contents of Bcl-2 and Bax in cardiomyocytes were detected by Western-blotting.Results: Compared with the ischemia-reperfusion model group, HE stain showed the changes of rats’ myocardial pathomorphology in the total flavones ofMappianthus iodoies groups was significantly improved; TTC stain showed that the myocardial infarct size of rats was reduced(P<0.05); ELISA analysis showed that the content of MDA, IL-6, TNF-α, CK and LDH of in serum were was significantly lower(P<0.05), while the content of SOD was higher(P<0.05);Western-blotting analysis showed that the expression of Bax proteins in the total flavones of Mappianthus iodoies group were significantly lower than those in the model group(P<0.05), the expression of Bcl-2 proteins in the total flavones of Mappianthus iodoies group were significantly higher than those in the model group(P<0.05). Meanwhile, compared with the shame group, HE stain showed the changes of rats’ myocardial pathomorphology in the model groups was significantly worse; TTC stain showed that the myocardial infarct size of rats was magnified(P<0.05); ELISA analysis showed that the content of MDA, IL-6,TNF-α, CK and LDH of in serum were significantly higher(P<0.05), while the content of SOD was lower(P<0.05); Western-blotting analysis showed that the expression of Bax proteins in the model group were significantly higher than those in the sham group(P<0.05), the expression of Bcl-2 proteins in the model group were significantly lower than those in the sham group(P<0.05).Conclusions: The total flavones of Mappianthus iodoies may have a protective effect in myocardial ischemic reperfused injury through anti-inflammatory,anti-oxidant and the inhibition of cell apoptosis.
