Purpose:We use rotenone in treatment of mouse macrophage cells RAW264.7 and observe the nuclear enrichment situation and the impact of murine macrophages from rotenone on P-mTOR signaling pathway, which explore the possible role of rotenone artery atherosclerosis. Methods:Use different concentrations of rotenone (0,0.05,0.1,0.3,0.5,1μM) in treatment of mouse macrophage cells RAW264.7 in vitro.Observe the nuclear enrichment situation and detected P-mTOR signaling pathway and its downstream protein P-S6K, P-S6 protein expression by Western blot. Results:After different concentrations of rotenone intervents mouse macrophages, we observe that the nuclear enrichment situation increases by increasing rotenone concentrations. And P-mTOR and its downstream proteins P-S6K, P-S6 protein expression are significantly increased. Conclusion:Studies have shown that rotenone by selective activation of mTOR signaling pathway in mice induced macrophage apoptosis to speculate rotenone.
