Objective:In this study,we detect the expression of PI3K signaling pathway related protein EGFR, PI3K and PTEN in human brain glioma.To analyze the relationship between the expression with the pathological grade, the clinical parameters of patients, and the correlation between these proteins,investigate the clinical significance,it was expected that our work could helpful to the clinical application of targeted therapy.Methods:50 available paraffin embedded samples collected from the First Affiliated Hospital of Kunming Medical University from January 2014 to June 2015. SP Immunohistochemical technique was applied to detect the expression of EGFR, PI3K and PTEN in 50 cases of specimens, the staining intensity and positive cell rate were recorded, using χ2 test to compare the expression differences in different pathological grade gliomas, gender, age and tumor size of each protein. Spearman rank correlation analysis was used to examine the correlation betweenEGFR, PI3K and PTEN.Results:In the present study,50 cases of glioma specimes.The positive rate of EGFR protein was 50%, the positive rate was 34.62% in the low-grade gliomas, the positive rate was 66.67% in the high-grade gliomas, and the difference was statistically significant (P< 0.05). The positive rate of PI3K protein was 60%, the positive rate was 46.15% in the low-grade gliomas, the positive rate was 75% in the high-grade gliomas, and the difference was statistically significant (P< 0.05). The positive rate of PTEN protein was 54%, the positive rate was 69.23% in the low-grade gliomas, the positive rate was 37.5% in the high-grade gliomas, and the difference was statistically significant (P< 0.05). The expressions of EGFR, PI3K and PTEN was no significant difference in different gender,age groups and tumor size(P>0.05).There was a positive correlation between the expression of EGFR and PI3K protein (r=0.327,P< 0.05),a negative correlation was fond between the expression of PTEN and EGFR or PI3K protein respectively.Conclusion(s):1. The expression level of EGFR and PI3K increased with the pathological grade of gliomas, the expression level of PTEN decreased with the pathological grade of gliomas。2. The expressions of EGFR, PI3K and PTEN was not related to gender,age groups(<50years or^50 years) and tumor size(<5cm or≥ 5cm).3.There was a positive correlation between the expression of EGFR and PI3K protein,EGFR overexpression may play an important role in the occurrence and development of glioma by activating PI3K signaling pathway.4. There was a negative correlation between the expression of PTEN and EGFR or PI3K protein respectively, Reveal that PTEN and PI3K signaling pathways may antagonize in brain glioma, PTEN expression decreased or deleted and EGFR overexpression together to promote the occurrence and malignant transformation of glioma through PI3K signaling pathway.
