| Objective:To study the expression of Fbln2 in glioma and adjacent tissues and its impacts on the biological behavior of glioma cell linesMethods:Real-time quantitative PCR (Real-time PCR) to detect differential expression of the fibulin-2 mRNA in three cases of glioma and peritumoral tissues research mRNA of Fibulin-2, the difference between each index factor compared by t test.Using the technology of RNA interference to construct specific gene silencing Fbln2 shRNA virus vector, packaging virs infected glioma cell lines U87mg cells, after the expression of fbln2 decreased by Real-time PCR and Western blot technique (western-blot) detection, by CCK-8 assay analysis U87mg cells proliferation, U87mg cells apoptosis was analyzed by flow cytometry, detect invasion and migration abilities of U87mg cells by transwell chamber; using gene overexpression technology, over-expression construct FBLN2 viral vectors, packaged virus.After U87mg cells infected with the virus, detected in the biological behavior of the above assay U87 cell proliferation, apoptosis, invasion and migration, etc. after FBLN2 increase gene expression by real-time PCR and Western technology, various factors differences between the index compared by t test.Result:1. FBLN2 mRNA levels in glioma tissues is significantly lower than adjacent tumor tissue (p<0.05, statistically significant difference); 2. After FBLN2 gene silencing U87mg cell proliferation relative to the control groups weakened whereas over-expression groups compared with the control ones U87mg proliferative enhancement (p<0.05. statistically significant difference); 3.Apoptosis after FBLN2 gene silencing on U87mg cell lines is enhanced, while there is no different in overexpression groups; 4. The invasion of U87mg cells is enhancement after FBLN2 gene silencing but it is decreased after overexpression(p<0.05 statistically significant difference); 5. The migration of U-87mg cells is enhancement after FBLN2 gene silencing but it is decreased after overexpression(p<0.05 statistically significant difference); (p<0.05 statistically significant difference)。Conclusion:Tumor tissues mRNA levels of fibulin-2 is significantly lower than the peritumoral tissues. In cell lines experiments, FBLN2 overexpression groups enhanced cell proliferation than interference groups, no significant changes in apoptosis changes. Invasion, migration:FBLN overexpression groups<FBLN2 normal groups<FBLN2 interference groups. This is reported in a previous non-metastatic solid tumors FBLN2 as a tumor suppressor gene, suggesting that fibulin-2 associated with the progression of glioma. |
