Lycoris is a perennial herbaceous plant with beautiful flowers and colour. Its ornamental value is high. Lycoris contain plenty of natural ingredients,lycoris and galanthamine was main object of adsorption, which have high medicinal value.Zeolite molecular sieve was main research objects in this research to experienced with the static and dynamic test on lycoris radiate and galanthamine for adsorption and elution, established detection method of lycoris and galanthamine and studied the specific purification method with zeolite molecular sieves on it, and compared with cationic resin and macroporous resin to experimental research. The specific methods and results are as follows:(1) The method of high performance liquid chromatography(HPLC) was used to determine lycorine and galanthamine. Test conditions are as follows: Syncronis C18(250 mm×4.6 mm,5 μm); flow rate of mobile phase was 1.0 m L/min; injection volume was 20μL; the column temperature was 30 ℃; mobile phase was 10:90 V(acetonitrile):V(phosphate), the phosphate was 2.72 g of potassium dihydrogen phosphate in 100 m Lwater with 1.4 m L triethylamine, adjusted the p H 2~3 with phosphoric acid, constant volume to 1L.(2) In contrast of performance, selected cationic resin D001, HD-8 and macroporous resin D101, AB-8 to compared with zeolite molecular sieve.Compared with macroporous resin and zeolite molecular sieve on static test, compared with cationic resin and zeolite molecular sieve on dynamic test. There were obvious advantages in adsorb and elute lycorine and galanthamine on static test with ZSM-5 H style(Si/Al=25) zeolite molecular sieve, than cationic resin and macroporous resin, adsorption rate achieved above 95.00%, the elution rate of lycoris and galathamine were 90.35% and 84.84%.Chemical properties might have more influence on D101 and AB-8 macroporous resin, the elution rate of galathamine were only 17.28% and 16.98%. Between both of D101 and AB-8, there was obvious advantage in adsorption and elution of lycoris dy AB-8, the rate of adsorption was above 95.00%, the rate of elution was 90.28%. In the dynamic test, the advantage of zeolite molecular sieve was not obvious, the gap between each sieve had certain influence on its performance.(3) The test compared to screen the ZSM-5, MCM-22 and 13 X zoelite molecular sieve, adsorbed and separated lycorine and galanthamine with H style, Na style and different ratio of silicate to aluminium. Extraction condition in test of Lycoris aurea was acid water extraction method and organic solvent extraction. Based on the rates of adsorption and elution, ZSM-5 H style Si/Al=25, was the best to separated lycorine and galanthamine, the optimum process as follows: the feed solution p H=4, adsorbing carrier was ZSM-5 zeolite molecular sieve, on the sample to the saturated at sampling rate 1 m L/min, then the zeolite molecular sieve was eluted at the rate of 1 m L/min with 1.0 mol/L ammonia water(contain 70% ethanol), in the elution dry material, lycorine was 40.09% and galanthamine was 19.93%. The content of lycoris and galathamine in extract liquid was 0.30% and 0.17%, the effect of purification was remarkable by compared.
