Background: Plasticity of visual system is one of the mechanisms of deprivation amblyopia. cPKC-γ as a neuron-specific protein, its role in visual developmental plasticity is below understood.Objective: 1.To investigate the dynamic expressions of cPKC-γ in visual cortex of normal mice. 2.Observe the effects of abnormal visual experience on cPKC-γ expression levels,then to explore the role of cPKC-γ in the plasticity of visual system.Methods: 1.Thirty-six clean C57BL/6 mice were collected.The mice were sacrificed at postnatal 7,14,21,28,35,42 days respectively to obtain the tissue samples of visual cortex.Expression levels of cPKC-γ in visual cotex following the ageing were quantitatively detected using Western blotting assay. 2.Twenty-four clean C57BL/6 mice were collected. Divided into two groups(P28 and P60). P28 groups were monocular deprived at P14. P60 groups were monocular deprived at 2 months. Each group take six mice to monocular deprive right eyes. The time of MD is 14 days. Then sacrifice them to obtain the tissue samples of visual cortex. Expression levels of cPKC-γ in visual cotex were quantitatively detected using Western blotting assay.Results: The expression of cPKC-γ in normal mice showed a dynamic increase with the ageing. cPKC-γ level in different days showing a significant difference.(Left,F=37.385,P<0.001;Right,F=21.098,P<0.001) MD have no significant effection on cPKC-γ expression of visual cortex in P28 groups and P60 groups.(P>0.05)Conclusions: cPKC-γ level in visual cortex presents a developmental change which correlated with the onset and decline of the critical period. After MD, no significant difference could be found on cPKC-γ expression. However, if there is a change in protein activity still needs a further research.
