| Objective:Ulcerative colitis(UC) of complex causes, the main disease in the intestine showed nonspecific inflammation. Dendritic cells(DC) plays an important role in the pathogenesis of the one and only UC. Toll like receptors(TLR) with different pathogenic microorganism combination of PAMP activates DC maturation and release ofcytokines participate. Modifie Pulsatilla Decoction in the treatment of ulcerative colitis(damp heat type), highlight the advantages of clear treatment. Through the study of Modifie Pulsatilla Decoction on the active ulcerative colitis(damp heat syndrome) patients, to observe the effects of changes in peripheral blood p DC and frequency function, and TLR9 Modified Pulsatilla Decoction on the expression of p DC. For the Modified Pulsatilla Decoction used in ulcerative colitis(damp heat type) to provide experimental basis for the treatment of.Methods:From July 2014 to June 2015 in the outpatient department and inpatient department of the second hospital of Shanxi Medical University active ulcerative colitis(damp heat type) in 40 cases, were randomly divided into A, B two group of 20 cases. Collect 20 cases of healthy subjects as healthy control group. The A group received routine therapy combined with oral Department of internal medicine beauty salad sustained-release granules of 1.0g/ times, 4 times / day. In the B group, A group based on the method of treatment with the concentration of Modifie Pulsatilla Decoction. The healthy control group without any intervention. At the start of the experiment and 12 weeks were collected from 3 groups of subjects with peripheral blood specimens. Using flow cytometry to detect peripheral blood p DC ratio and TLR9 expression; purification of peripheral blood p DC by magnetic bead sorting method in super clean bench, Cp G-ODN2216 to 24 hours after the purification of p DC activated by enzyme-linked immunosorbent assay(ELISA) detection of IFN-α concentration method.Results:The results showed that: before treatment, the p DC of PBMC in peripheral blood of healthy control group was 0.43%±0.16%, which was significantly higher than that of patients in group A was 0.04% ± 0.02% and 0.02% in group B,(P<0.05). After 1 courses of treatment, the PBMC ratio of peripheral blood p DC in patients with A group was0.22% ± 0.09%, lower than that in group B 0.37% ± 0.09% and healthy control group0.44% ± 0.16%(P<0.05). Before treatment, the healthy control group p DC expression in peripheral blood of the mean fluorescence intensity of TLR9 61.93 ± 8.60, was significantly higher than that in A group were 41.90 ± 8.67 and 41.87 ± 8.84 in the B group(P<0.05). After 1 courses of treatment, patients in group A p DC expression in peripheral blood of the mean fluorescence intensity of TLR9 is 51.87 ± 8.91, 62.13 ±8.72 lower than that of B group and healthy control group 62.03 ± 8.85(P<0.05). The healthy control group before treatment, peripheral blood p DC IFN-α secretion level was(622.87 ± 86.02) pg/m L, significantly higher than in group A(321.57 ± 84.99) pg/m L and B group were(322.33 ± 85.43) pg/m L,(P<0.05). After 1 courses of treatment, group A patients with peripheral blood p DC IFN-α secretion level was(472.77 ± 85.79) pg/m L lower than that of group B(621.40 ± 86.25) pg/m L and healthy control group(621.27 ±85.24) pg/m L(P<0.05).Conclusion:Ulcerative colitis and plasmacytoid dendritic cell frequency and function of plasmacytoid dendritic cell is closely related to the expression level of TLR9. Modifie Pulsatilla Decoction can improve the active ulcerative colitis(damp heat type) and the frequency of p DC in peripheral blood of patients with up-regulated expression of TLR9,p DC and enhanced IFN-α secretion ability. And the Modified Pulsatilla decoction combined with the beauty of salad sustained-release granule in treating ulcerative colitis(damp heat type) than used alone in the treatment of salad beauty sustained-release granule effect is good.
