Objective:To investigate the association of the HRCT features and expression of serum TNF-αand PDGF in the coal worker’s pneumoconiosis(CWP) rabbits model.Methods:1. Animal experiment selected species(healthy Japanese male) in rabbits with 40, in accordance with the principle of random assignment, divided into 5 groups, 8 rats in each group, corresponding to the 2 weeks, 1 month, 2 months, 3 months, 5 month staging and rabbits in each group choose 3 only as control group, and as the experimental group, control group rabbits were a total of 15, rabbits in the experimental group totaling 25 only. Making coal dust concentration of 100 mg / ml of suspension, using 4% pentobarbital anesthetized rabbits, in under the guidance of the laryngoscope for neonate respectively on rabbits successful tracheal intubation and perfusion of 2ml / kg in accordance with the standard, the experimental group rabbits trachea injection coal dust suspension and control group rabbit tracheal injection dose of normal saline.2. In the 2 weeks, 1 month, 2 months, 3 months, 5 month, when the group rabbit row corresponding to the high resolution CT scan, careful observation of all stages of each group rabbits pulmonary fibrosis imaging, and carefully recorded.3. In the 2 weeks, 1 month, 2 months, 3 months, 5 month, the white rabbit ear after scanning by venous blood collection, using enzyme-linked immunosorbent assay to detect serum TNF- alpha, PDGF unified content, and carefully recorded.4. SPSS 20.0 software was used for data analysis, independent samples t test was used to compare the mean value between the two groups. In the experimental group each stage were compared using analysis of variance, the pairwise compared with Q test. Correlation analysis using Spearman rank correlation analysis and rank sum test. P< 0.05, there were statistically significant differences.Results:The control group rabbit lung HRCT findings: each period of each period of rabbit lung texture clear, natural walking, no abnormal density. Rabbit lung in experimental group 2weeks: mainly for lung slightly high density shadow, fuzzy boundaries, showed ground glass density; January: showed ground glass opacities increased, interlobular septal thickening;February: increased fibrosis image, around bronchovascular bundle often accompanied by fiber hyperplasia, edema and inflammatory exudation; March: the further development of the disease, the visible coal nodule formation involving the pleura, honeycomb change. May:late disease, lung fibrosis and bronchiolectasis, extensive cellular image formation. The level of TNF-α in serum of CWP rabbits was lower, but the level of PDGF was higher in comparison with those of the control(P<0.05, respectively).Moreover, with the pulmonary lesions aggravating, serum TNF-α and PDGF decreased gradually. TNF-α expression level was positively related to PDGF(P<0.05). TNF-α、PDGF expression levels were negatively related to the severity of pulmonary interstitial fibrosis(P<0.05)Conclusion:Pulmonary fibrosis imaging is a typical performance of rabbit CWP periods can be observed in the HRCT image to prove, rabbit CWP model preparation is successful. The expression levels of TNF-α and PDGF in serum maybe related to the severity of pulmonary interstitial fibrosis, TNF-α and PDGF play an important role.
