Acanthopanax trifoliatus (Linn.)Merr.is a kind of medicine plant belonging to Acanthopanax. The root, root bark, burgeon with leaf or whole plant used as medicinal materials.It has the effects of "clearing away heat and toxic material" "blood circulation and relaxing muscles" and "wind-dispersing and damp-inhibiting". And mainly used to treat cold and fever, pharyngalgia, cough, chest distress, etc. At the same time, the burgeonand leaves are eaten as a vegetable in the folk, can be used to fry and cook. Moreover, tea leaves can be made.So, the study on it in gradually increased in recent years.The use of Acanthopanax trifoliatus (Linn.)Merr.stay in folk for a long time, and not only rich in resources, its effect also is notable. But no one has expanded on its comprehensive study on pharmacognosy research. To made the character of pharmacognosy clear, ensure the safety and efficacy of the medicinal, and establish an overall、effective system of quality assessmen. Therefore, we carried out the study on pharmacognosy from characteristic identification, microscopic identification, physical and chemical identification, leaf morphological and venation pattern identification and DNA barcoding identification.Firstly, it has obviouscharacteristic. And microscopic identification showed that cross-section of the root, stem and leaf there were resin canals. In addition, the cells of the anterior ray were aggrandizement in the root cross-section, and like a megaphone. 2~4 layers collenchymas were observed beneath the epidermis of stems cross-section, and there were calcium oxalate crystals scattered. Non-glandular hairs and flat shaft pores were common in leaf abaxial epidermis. The main vein of the cross-section of leaf has collateral type vascular bundles. Stone cells, calcium oxalate crystals, sclerenchymatous cells, fragment of resin canals, fibers, cork cells, threaded catheter and starch grains in the powder.Second, the thin-layer chromatography(TLC) was used to respectively build the quality control method for rutin and chlorogenic acid. And the UV absorption was built by UASLG method take on a strong characteristic in quality.Third, the identification characters of morphological and venation pattern of Acanthopanax trifoliatus (Linn.) Merr.andAcanthopanax gracilistylus W. W. Smith were distinct. With the characters of situation at the base of barbed spines on petiole, the widest position, the number of sencondary veins per side, the dichotomously branching times of sencondary veins, the symmetric characteristics about meshes of tertiary veins, and the looped phenomenon in the lower part of the leaf margin about it, the level of ultimate venations and the dichotomously branching times, which can be used to distinguish them.Fouth, DNA barcoding base on ITS2 sequence not only provided that intrinsic molecular information of Acanthopanax trifoliatus (Linn.) Merr., but also further improved the pharmacognosy research on it. This molecular biological methods provided an effective method for identification research of Acanthopanax trifoliatus (Linn.) Merr.According to the subject study, a new thought for comprehensive subject results to improve the quality standard, and it can provide a reliable basis for the future identification, development and utilization.
