Objective To research on antimalarial compound extract of CQ and preparation process optimization research, to provide experimental data for new drug application.Method1 Establish the pharmacodynamics of microbial and chemical composition analysis of CQ compound extracts then use the two methods to evaluate the pharmacodynamics of compound extract of CQ.2 Optimize extraction process of CQ extract by chemical method, respectively on compound CQ, solvent, extraction solvent extraction and extraction times four factors studied. Extracted with alcohol consumption, time, and number three, three levels, and L9(34) orthogonal design to optimize extraction process, select the best extraction process I.Based on the optimized extraction process of I, combined with the demand for industrial production of saving metal material and ultimately determine the optimum extraction process of compound CQ II. According to the extraction II prepare 3 batches of compound extract of CQ, validation of the process. Preparation of compound extracts of CQ’s best.3 Research on compound extracts of CQ’s best, including high performance liquid chromatography study, study on thin-layer chromatography and pharmacodynamics.Results1 Ge Hong extracting method for artemisinin extraction rate is 23.11%;Ether Soxhlet extraction method of artemisinin only 13.19%, compared to 9.01% infusion;To compound other solvent filter the CQ, extraction rate is petroleum ether group for 48.76%, n-butyl alcohol group is 34.34%, chloroform was 27.78%,75% alcohol group is 70.03%,acetic acid ethyl ester group is 12.78%,acetone was 11.72%. Extraction solvent screening result,best ethanol extraction rate groups, significantly higher than the other groups, including ether group.2 Factors and orthogonal optimization to be 7 times 85% ethanol extracts of compound three times, every 30 minutes, artemisinin content to 11.25mg/g, the extraction rate of92.20%.3 Compound CQ extract by high performance liquid chromatography method for the determination of Arteannuin content for 10.42mg/g;Thin-layer chromatography results CQ extracts tested speck in the spectrum, in comparison with artemisinin Speck on the location of the corresponding, same color spots;Efficacy results in Kunming strain of mice infected with Plasmodium Burt K173 negative control inhibition rate of 73.70%.Conclusion1 Chemical evaluation of established methods and assessment methods can be used to evaluate the pharmacodynamics of compound extract of CQ.2 Anti-malarial compound CQ extraction method, that is 7 times 85% ethanol extracts of compound three times, every 30 minutes, artemisinin extraction rates are higher than diethyl ether extraction, petroleum ether extraction and extraction of Ge Hong; Plasmodium Burt’s strong resistance to petroleum ether, chloroform and other groups providing technology can provide the basis for antimalarial drugs declared.
