Agaricus bisporus was also known as mushrooms and white mushrooms, which was also called button mushroom in Europe and the United States.It growed in grass in low temperature and widely distributed.People knew and eat Agaricus bisporus in ancient Greece,meanwhile it was artificial cultivate in early 17 th century. Artificial cultivation technology of Agaricus bisporus has become more mature. To explore more strains, this paper studied the Agaricus bisporus from Tibetan and domesticated it to develop new varieties and protect the germplasm resources of this species and enrich the Chinese wild Agaricus bisporus mushroom germplasm resources database.Because the strain were collected from high-altitude area and the local climate was mountain plateau climate, so its qualities were conducive to adjust the structure of the varieties of the strain and selection by improvement domestication and cultivation.(1)In wild Agaricus bisporus domestication and cultivation experiment, According to the mycelium growth rate of solid spawn and liquid spawn to choose the best female parent culture medium and mother seed culture medium, and the cultivation experiment was carried out.The results indicated that the best solid female parent culture medium(pH unadjusted) consisted of(/L) potato 200 g,glucose 20 g, peptone 3g, potassium dihydrogen phosphate 1g, magnesium sulphate 0.5g, agar 20 g and the best liquid culture medium concluded(g/L)Soybean meal31.15,corn starch31.13,peptone3.16 and mother seed of culture medium consisted of wheat 93%,cow dung powder5%,gypsum1%,lime1%.The growth cycle of mother seed by liquid spawn was 8days earlier than solid spawn,so the liquid spawn was more dominant than the solid spawn,but the difference between them was not obvious on cultivation.(2)Agaricus bisporus fruit body polysaccharide and fermentation polysaccharide(intracellular polysaccharides and extracellular polysaccharides)regulated T lymphocyte subsets CD3+CD4 +, CD3+CD8+ content in the normal immune regulation experiment. At the same time, the ratio of CD4+/CD8+ was dose dependent increase, also in the regulation of Th1 and Th2 cells, cytokine IL-2,IL-4, TNF-α, IFN-γsecretion quantity.They jointly enhanced the body immunity.(3)In immunosuppressive experiment, Agaricus bisporus fruit body polysaccharide and fermentation of polysaccharide each group was dose dependent recovery of spleen lymphocyte proliferation and peritoneal macrophages phagocytosis in vitro. The cytokine IL-2, IL-6, TNF-α,IgG increased, and IL-10 and IL-17 had significantly decreased, indicating that Agaricus bispo-rus polysaccharide and fermentation polysaccharide could reverse cyclophosphamide induced immunosuppression and adjust the balance of immune.(4)In CCl4 induced liver injury experiment,The polysaccharide of fruit body and fermentation polysaccharides could reduce the levels of ALT,AST,GGT,MDA and raise SOD, GSH index.Meanwhile they adjusted the TGF-β1/ Smad signal pathway which TGF-β1 and Smad3 protein expression decreased and Smad7 protein expression increased, and were consistent with immunohistochemical experiments results.
