Evaluating The Effect Of Duhuojisheng Decoction Through The Changes Of Cartilage Metabolism Markers In KOA

Objective:(1)To detect the COMP and aggrecan content in the serum of patients with knee-osteoarthritis(KOA), and to investigate the correlation factors.(2) To study the change of COMP, aggrecan and total knee magnetic resonance imaging(WORMS) after the treatment of Duhuo Jisheng decoction on patients with KOA, and to observe the effect of Duhuo Jisheng decoction on KOA, and discussion on the mechanism and material basis of the treatment of KOA, to provide a theoretical basis for the modern clinical application of traditional Chinese medicine recipe.Method:(1) COMP and aggrecan in the serum of 100 patients with knee-osteoarthritis(KOA) and 30 healthy volunteers were measured by ELISA. The correlative factors for COMP and aggrecan levels were tested by linear regression analysis.(2) Totally 80 elderly patients with KOA were randomly divided intotwo groups(each contains 40 cases).The control group additionally took Hydrochloric acid glucosamine capsules, and the treatment group was given Duhuo Jisheng decoction besides taking Hydrochloric acid glucosamine capsules. After 8 weeks, WOMAC scores of the two groups were determined, the imaging of KOA was assessed by WORMS,COMP and aggrecan in serum were detected by enzyme linked immunosorbent assay(ELISA).Results:(1) The levels of COMP and aggrecan in KOA were significantly higher than that in the healthy volunteers(P<0.05). The COMP and aggrecan levels in serum were positively correlated with age, body mass index(BMI), WOMAC scores and X-ray.(2) The effective rate in the treatment group was 86.84%, higher than control group(71.79%),the difference was statistically significant(P<0.05).There were significant differences in WOMAC scores between the before and after treatment, so as that between the two groups(P<0.05).The COMP and aggrecan concentration and WORMS decreased in both of the 2 groups after the treatment,and more significantly in the treatment group(P<0.05).Conclusion:(1) In the KOA, the levels of COMP and aggrecan in the serum are elevated the one of normal.Moreover, the elevated levels positivelycorrelated with clinical disease parameters and radiological joint damage.(2) Duhuo Jisheng decoction can protect articular cartilage by reducing the concentration of COMP and aggrecan,and improve clinical efficacy and decrease WORMS of elderly KOA.
