| Objective: This study was designed to investigate the effects of Curcumin on apoEã€RXR and LXR on cerebral ischemia-reperfusion injury in rats and in Primary Cultured Astrocytes Under Oxygen-glucose deprivation/Reoxygenation respectively.Methods: in vivo experiments, using Plug-line method make focal cerebral ischemia and reperfusion of middle cerebral artery occlusion model of rat. Adult male SD rats were randomly divided into five groups: sham group(SH); ischemia-reperfusion group(IR); Curcumin- treatment groups: a dose of 100 mg/kg; Curcumin- treatment groups: a dose of 300 mg/kg;Nimodipine positive control group:a dose of 10 mg/kg. Make Neurological deficit scores estimate, Measure Brain water content, Using Western blotting(Western Blot) to detect the expression of apoEã€LXR and RXR, Make HE to observe morphological changes of brain tissue,Make TUNEL to detect neurons apoptosis in rat hippocampal CA1 at 24 h. In vitro experiments, Primary cultured astrocytes, using the way of oxygen-glucose deprivation/reoxygenation make cerebral ischemia-reperfusion model in vitro.Through the expression of glial fibrillary acidic protein(GFAP) by using immunofluorescence to identify astrocytes. In this study, the cultured cells were divided into seven groups: normal control group; the model group; curcumin group: 2.5 umol/L, 5 umol/L, 10 umol/L, 20 umol/L; nimodipine group: 0.5 umol/L. Viability and cytotoxicity of cells to be assessed by MTT and LDH. Western blotting was used to detect the expression of apo E, RXR, LXR.Results: in vivo experiments,Curcumin in the dose of 100 mg/kg and 300 mg/kg can improve the nerve damage symptoms of MCAO rat; Reduce brain water content; Relieve brain morphological changes and neuronal apoptosis; Increase the expression of apo Eã€LXR and RXR. Especially,the treatment effect of Curcumin 300 mg/kg groups was the most significant. In vitro experiments, Curcumin can significantly increased cell viability and reduced LDH release. The expression of apo E, RXR, LXR was increased. In it, 20 umol/L were most obvious and the lowest dose 2.5 umol/L was almost ineffective.Conclusions: Curcumin has a protective effect for Cerebral ischemia and reperfusion and Primary Cultured Astrocytes under Oxygen-glucose Deprivation/Reoxygenation, This protective effect may be regulated by the expression of apoE LXR, RXR arising. |
