| Objectives1. By analyzing the expression of NM23-H2 protein and STAT3,P-STAT3 in the tissue of colorectal adenocarcinoma, lymph node metastasis and its relationship with clinical pathological characteristics, preliminary discussion of the role in colorectal adenocarcinoma occurrence, development and metastasis.2.Through analysis the expression of NM23-H2 proteins and STAT3, P-STAT3 in colorectal adenocarcinoma lymph node metastasis and the prognosis of patients with statistical, preliminary discussion the clinical significance in the clinical prognosis of patients with colorectal adenocarcinoma.3.Preliminary analysis the correlation of NM23-H2 protein expression and the JAK-STAT signal pathway activation state in colorectal adenocarcinoma, providing the basis for the further study colorectal adenocarcinoma molecular mechanism of occurrence, development and metastasis.Methodone hundred and twenty-four cases of cancer surgery patients information was collected nearly 5 years in lanzhou military region general hospital, the patients colorectal carcinoma tissue speciens of paraffin wax was obtained from the block garage in the Department of pathology. Included 42 patients had lymph node metastases, and obtained 20 cases wax block of adjacent to the corresponding colorectal tissue around, adjacent to carcinoma 15 cm away, preparation instrument using tissue microarray and tissue microarray. Using the immunohistochemical method to dye the adenocarcinoma tissue, lymph nodes metastasis tissue, tissue adjacent to carcinoma respectively.The expression of NM23-H2 protein and transcription factors STAT3, P-STAT3 was observed under light microscope. Analysis of its pathology meaning in all three groups, and the relationship with clinical pathological parameters, analysis the relationship between NM23-H2 protein, the transcription factors STAT3, P-STAT3 and the prognosis of patients. All the data was processed using SPSS, P<0.05 showed the difference was significant.Results1.The expression of NM23-H2 and STAT3, P-STAT3 in colorectalAdenocarcinoma, metastasis lymph node tissue, the tissue adjacent to carcinoma are different. In metastasis lymph node tissue and metastatic colorectal adenocarcinoma tissues the expression of NM23-H2, STAT3 and P-STAT3 also have differences. In colorectal adenocarcinoma tissue and metastatic lymph nodes in the expression of NM23-H2 and STAT3, P-STAT3 have negative correlation, the difference is statistically significant.2.In colorectal adenocarcinoma tissues, the expression of NM23-H2 proteins and STAT3, P-STAT3 is relevant with colorectal adenocarcinoma tissue pathological grade and TNM classification, whether lymph node metastasis, difference is statistically significant.3.Single factor survival analysis found that patients with colorectal adenocarcinoma of overall survival, progression-free survival is relevant with pathological classification of colorectal adenocarcinoma, the expression of NM23-H2,STAT3 and P-STAT3, TNM staging of colorectal adenocarcinoma, if metastatic lymph nodes.Multiariable analysis found that the patient’s overall survival, progression-free survival is relevant with the expression of P-STAT3 in colorectal adenocarcinoma tissues and TNM, has nothing to do with other pathological parameters.Conclusion1.NM23-H2 gene may play a potential inhibitory effect on the occurrence and metastasis of colorectal adenocarcinoma.2.JAK-STAT may be have a potential role in promoting on the occurrence and metastasis of colorectal adenocarcinoma.3.P-STAT3 can be used as potential molecular targets in the clinical treatment of colorectal adenocarcinoma and prognosis of patients with evaluation indicators. |
