More and more scientists have been drawn to the marine-derived fungi because of their special living environment and metabolic way. As a large number of novel and bio-active compunds have been isolated from the secondary metabolites of marine-derived fungi, they have been a large resource library of lead drug.The thesis included three charpters. The research progress of secondary metabolites from marine-derived fungi was introduced. The studies on the secondary metabolites of two mangrove fungi(#008, #035) collected from the South China Sea were carried out the first time. 21 compounds were isolated from the culture of these fungi through chromatographic separation. Their structures were deduced from the data of 1H NMR, 13 C NMR, 2D NMR, IR and FABMS, these compounds related to multiple struture types including Steroid, Peptide, fatty acid, and so on.In charpter one, the research progress of the novel compounds, which displayed antitumor, antimicrobial, enzyme inhibition, antimalarial and other bioactivities, from marine micro-organisms in recent years were summarized, including Alkaloid, Polysaccharide, Polyketide, Peptide, Steroid, Terpenoids, Piperazine, Lactones, Phenols, and so on.In the second chapter, the secondary metabolites of the mangrove endophytic fungus #008(Alternaria tenuissima) were studied. 12 componds were isolated through various technical, such as fermentation cultivation, extraction, chromatofraphic, recrystallization, and so on. These compounds were identified by spectrum analysis(silica gel column chromatography, gel fitration, HPLC, Semi-preparative liquid chromatography) and physical-chemical constant comparing with literature. Compounds include(2-1) β-sitosterol,(2-2) cerevisterol,(2-3) 5, 8-Epidioxy-5α, 8α-ergosta-6, 9-22E-tien-3β-ol,(2-4)(22E)-5a, 8a-epidioxyergosta-6, 22-dien-3b-ol,(2-5)(Z)-10-eicosenoic acid,(2-6) Stearate,(2-7) Tentoxin,(2-8)(22E, 24R)-23-Methylergosta-7, 22-diene-3?, 5α,6β-triol,(2-9) methyl oleate,(2-10) β-sitosteryl galactoside,(2-11) cyclo-(L-Phe-L-Tyr), 3-benzyl-6-(4-hydroxybenzyl) piperazine-2, 5-dione,(2-12) 2, 6, 8-trihydroxy-4-methylphenanthren-9(10H)-one.In the third chapter, the secondary metabolites of the mangrove endophytic fungus #035(Pestalotiopsis) were studied. 12 componds were isolated through various tschnical, such as fermentation cultivation, extraction, chromatofraphic, recrystallization, and so on. These compounds were identified by spectrum analysis(silica gel column chromatography, gel fitration, HPLC, Semi-preparative liquid chromatography) and physical-chemical constant comparing with literature. Compounds include(3-1) Thioacetic anhydride,(3-2) 7-Hydroxy-5-methoxy-4, 6-dimethylphthalid,(3-3)(22E)-5a, 8a-epidioxyergosta-6, 22-dien-3b-ol,(3-4) citreorosein,(3-5) 4-hydroxyphenylacetic acid,(3-6) 4-(2-hydroxyethyl) phenol,(3-7) 5-hydroxymethyl furfural,(3-8) succinic acid,(3-9) Cyclo(Phe-Ala),(3-10) palmitic acid,(3-11) β-sitosterol,(3-12) Stearate.
