Objective:Observe the influence of Chinese herb preparation YuHong oil gauze on traumatized wound healing model,and explore the molecular mechanisms of its action to promote wound healing.Method: Divided 96 SD rats into A(Control group), B(Model group),C(Positive control group),D(YuHong oil gauze group) four groups by randomization method,each group of24.Establishing trauma model by surgery for the rats of B,C,D three groups.Different groups of rats treated the wound dressing and observe the wound tissue changes very day,record healing time.Randomly 8 rats of each group respectively in the dressing 4th day and the 10 th day,heart blood and excised wound tissue.Observed the wound morphology,inflammatory cells,granulation tissue,blood vessels,fiber changes and wound healing time under optical microscope for each group rats.Detect the serum VEGF, IL-6 and TGF-β levels by ELISA;Detect the protein levels of VEGF,IL-6 and TGF-βin wound tissue by Immunohistochemistry;RT-PCR detection of wound tissue mRNA expression of VEGF,IL-6 and TGF-β.Result: ①Gross observation the wound after dressing,visible the wound of B,C,D groups rats were tissue edema, exudation gradually reduced,increased scab tissue and wound shrink over time.C,D groups change significantly than group B,the wound healing time was significantly shortened.②HE staining show different time points after dressing inflammatory cell proliferation proliferation significantly in the wound tissues of rats in group B than group C,D,and gradually reduced over time.B,C,D groups rat wound healing capillaries, fibroblasts and granulation tissue gradually increased,C,D significantly increased than group B in the same time.③SP immunohistochemical and RT-PCR detected VEGF,TGF-β and IL-6expression results at different time points after dressing the wound tissue of rats display:C,D groups TGF-β and VEGF expression in rat wound tissues than in group B,the difference was statistically significant(P<0.05),and with the passage of healing time is a growing trend;C,D two drugs on the expression of wound tissue in a rat model of IL-6 has significant control effect,compared with group B, the difference was statistically significant(P<0.05).④After dressing rats in each group at different time points in serum VEGF,IL-6 and TGF-β content ofthe statistics:After 4 days compared the serum of TGF-β and VEGF content each groups,the difference was not statistically significant(P>0.05);After dressing 10 days C,D groups in serum TGF-β and VEGF were higher than A,B groups,the difference was statistically significant(P<0.0 5);Different time points after dressing C,D groups serum IL-6 content of rats with A,and B group,the differences were statistically significant(P <0.05).Conclusion: ①YuHong oil gauze can effectively reduce wound edema and exudate,promote wound fibers and granulation, shorten healing time.②Jade marked gauze can be controll the wound inflammation, increase wound angiogenesis and granulation, promote wound fast and effective healing by intervene the IL-6,VEGF and TGF-βexpression in wound tissue of traumatic model.
