Objective: To study the changes of CD83 in dendritic cells(DCs), Th17 and Treg in mice with chronic obstructive pulmonary disease(COPD), and to further explore the immune pathogenesis of COPD.Methods: The C57BL/6 mice of SPF grade were divided into healthy control group(n=10) and COPD model group(n=16). Exposure to cigarette-smoking was used to establish the mice model of COPD.At 28 days of the experiment, the mice in two groups were sacrificed and the peripheral blood, bronchoalveolar lavage fluid(BALF) and lung tissue samples were collected. The lung tissue were investigated by HE staining. The changes of CD83, Th17 and Treg in peripheral blood, BALF and lung tissue were detected by flow cytometry.Results: 1. The lung tissue in the COPD model group were stained by HE showed significant pulmonary inflammation and formation of emphysema;2. Compared with the healthy control group, the percentage of CD83 was decreased in the peripheral blood, BALF and lung tissue of COPD model group(P<0.05);3. The percentage of Th17 in the peripheral blood, BALF and lung tissue of COPD model group was higher than that in the healthy control group(P<0.05);4. The percentage of Treg was decreased in the peripheral blood, BALF and lung tissue of COPD model group compared with that in the healthy control group(P<0.05);5. The ratio of Th17/Treg in the peripheral blood, BALF and lung tissue of COPD model group was higher than that in the healthy control group(P<0.05).Conclusion: Cigarette smoke could suppress the normal DCs maturation and CD83+DCs may play an important role in the pathogenesis of COPD;Cigarette smoke could lead to the imbalance of Th17/Treg, the imbalance of Th17/Treg may be involved in the immune pathogenesis of COPD.
