Cacumen Platycladi, the typical used Chinese medicine, is the dry branch and leaf of Platycladus orientalis(L.)Franco. It has performed as the key medicine for blood-cooling and hemostasis, as well as relieving cough and removing the phlegm. Though it has good effect in hemostatic, most medical practitioners are used to choosing Cacumen Platycladi Carbonisatum, based on the theory of "Black beats Red."Cacumen Platycladi Carbonisatum is the research object of the essay, which concludes 5 parts: literature review, hemostatic mechanism, processing procedure, quality standard and fingerprint.1. Literature reviewWe have reviewed relative ancient and modern documents, collate many research results and discussion, and summarize a review about the historical change of processing mechanism of Cacumen Platycladi from the ancient documents, the processing and clinical experience, chemical compositions, qualitystandard, pharm acological and clinical applications.2. Hemostatic mechanismWe choose the BT and CT as indexs, do the typical pharmacological experiment. The results implies that the flavonoids part is the effective part of hemostasia. Meanwhile, we compare the flavonoids part, the tannin part between raw product and carbonized product. It turns out that after carbonized, the amount of flavonoids reduces 26.57%, tannin reduces 48.15%, and the quercetin increases 37.27%, compares to raw product. So we infer that the change of content and component of flavonoids part is closely related to the better effects in hemostatic.3. Processing procedureTaking quercetin as the investigated index, we use intelligent stir-frying machine of drum type to screen the best processing technology by orthogonal test. The result implies that there is huge difference between laboratory and factory. The best parameters is 250℃, processing 6min. The technology is steady, we take it for pilot test to produce 10 batches of samples. The yield rate is higher than 85%. 4. Quality StandardWe follow the method of pharmacopoeia to do qualitative, including microscopic, physicochemical and thin layer identification of optimal slices, as well as the determination of water, ash content, alcohol extracts, heavy metal, and index components content. We formulate the draft that, the content of water must limited in 1.0%; the ash content must limited in 14.5%; the acid insoluble ash content must limited in 4.0%; the water extract is no less than 14.5%; the 50% alcohol extract is no less than 13.5; the amount of flavonoids is no less than 0.035%; the amount of quercetin is no less than 0.01%.5. FingerprintBy using the UPLC method, we get some studies on fingerprinting. The chromatographic column was Acquity UPLC BEH C18 (2.1mm×50inm,1.7μm), the mobile phase was Methanol and 1% formic acid water, the detection wavelength was 375nm, the flow rate was 0.2ml/min. Compare to HPLC,UPLC has many advantages. We suggest it as a tool to control the quality for Cacumen Platycladi Carbonisatum.
