In order to adapt to the special ecologic environment, many fungi in the deep sea ocean have captured the metabolic path ways to survive from the servere conditions like high presure, high salt density and poor nutrition, during which many compounds with special structures and functions are produced, and many are new compounds.FS50, the strain used in this study, was believed to have relately good activity and abundant compounds in the extract of PDA culture and mycelia in our preliminary experiment. DNA identification of FS50showed that it belonged to Penicillium sclerotiorum, which was conformed by scanning electron microscope.The extracts of FS50from the the culture and mycelia were obtained respectively by large scale fermentation in the PDB. From the culture extract five compounds were isolated and the mycelia extract eight by repeated columnchromatography on silica gel, C18reverse phase, Sephadex LH-20, preparative thin layer chromatography, HPLC and recrystallization. The structures of these compounds were determined by the means of spectroscopic techniques including1H-NMR,13C-NMR, ESI-MS, EI-MS DEPT135, HSQC,1H1H-COSY, and HMBC. The compounds were sclerotiorin (SC-01),4-hydroxyacetophenone (SC-02), pterocarpol (SC-03), penicilazaphilone B (SC-04),3-carboxy-8-hydroxy-6-methoxy-1-oxo-1H-2-benzopyran (SC-05), pterocarpol (SC-06), isochromophilone ? (SC-07), glycerol monolinoleate (SC-08),(3b,5a,6b,22E)-6-methoxyergosta-7,22-diene-3,5-diol (SC-09), tyrosol (SC-10), di-(2-ethyl)-hexylphthalate (SC-11), cerevisterol (SC-12), sclerotioramine (SC-13), among them SC-03from the culture and SC-06from the mycelia were the same compounds. SC-08, SC-09, SC-10, SC-11, SC-12and SC-13were isolated for the first time from the species of P. sclerotiorum, while SC-03(SC-06) and SC-05were isolated for the first time from the genus of Penicillium species. SC-04was isolated for the second time.Doctor Zeeck's OSMAC strategy (one strain many compounds) was employed in this experiment by altering the culture for FS50. Rice solid medium was used to culture the fungus from which seven compounds had been obtained. Their structures were dertemined with spectroscopic techniques. The compounds are4-methyl-5,6-dihydropyren-2-one (GSC-01),5-hydroxymethylfurfural (GSC-02),3-carboxy-8-hydroxy-6-methoxy-1-oxo-1H-2-benzopyran (GSC-03same as SC-05), daucosterol (GSC-04), sclerotiorin (GSC-05same as SC-01), butyl6,8-dihydroxy-1-oxo-1H-isochromene-3-carboxylate (GSC-07), among them GSC-01and GSC-04were isolated for the first time from the species P. Sclerotiorum GSC-03(SC-05), GSC-02and GSC-06were isolated for the first time from the genus of Penicillium species, and GSC-06was isolated for the second time according to the literatures, while GSC-07was identified as a new isocoumarin compound.Prelimilary antibacterial activity to Escherichiacoli, Staphylococcus aureus and cytotoxic activity to SF-268, MCF-7, NCI-H460and HepG-2of some compounds were evaluated with Resazurin, MTT and SRB methods. Unfortunately, no distinguished antibacterial or cytotoxic activities were observed.
