This thesis were researched in the classification and molecular identification of fungus specimens collected from hainan,jiangxi provinces of live or dead leaves specimens and the section saved in the Reference Collection of Frest Fungi of Anhui Agricultural University(AAUF).This paper established a comprehensive system research from the aspects of fungus morphological anatomy,physiology,ecology,biology and individual development by the modern fungus classification technology.With the development of modern molecular techniques,the paper constructed the phylogenetic trees against discomycetes division(Rhytsmataceae Chevall.)based on the sequences of rDNA-ITS,28 sr DNA and two-gene combined sequences respectively.And it's discussed the division between genera,the genus,interspecific and intraspecific genetic relationship and genetic diversity.The Rhytismataceae samples were stdutied on the main basis the principle of classification systems puted forward by Darker(1976),Cannon & Minter(1983,1986),Johnston(1986).Thirteen species in six genera of Rhytismataceae were identified.They included five new species,Coccomyces chuniae,Terriera fici,T.elongata,T.rosae,T.sparsa.And among them,C.crassissimus,C.sinensis,L.australe,L.heterocladum,L.pini-excelsae,M.desmazieri,M.chinensis,R.himalense were already known in china.This article provided detailed description,discussion,habital,line drawings and photographs for new species.And briefly described and analyzed of morphologic features were given for the other species.The extraction of genomic DNA of strains was used by CTAB method,identified and purified sequences and look for suitable sequence in GeBank.Phylogenetic trees were established used Leotiia lubrica as outgroup based on 28 srDNA,ITS-rDNA and two-gene combined sequences.The analysis of phylogenetic trees showed: The significant features among ascoma fungal genera was the shape and openning mechanism of ascomata and the shape of ascospore,so buried depth and paraphysis characteristics of ascoma is not the key characteristics between genera.The significant factors in Interspecific relationship were the depth and external shape of the ascocarp,opening mechanism,the character of paraphysis and host speciesis.The classification of features in the specis relationship were host species(herbs,conifer and broadleaf trees)and the shape of the ascospore.However,there is insufficient in the process of analysis.For all this,the main reasonable basis fungi classification is still traditional morphology method.The scientific and reasonable method will be established combining morphology,phylogeny,genetic physiology and biochemistry and nutritional componentswith molecular biotechnology of fungi.
