Phosphorus is an important plant nutrient,and it regulates entire metabolic processes.However,the content of phosphate available to plants in soil is very low,and it often cannot meet the requirements of plants.Therefore,the available phosphate supply in soil and the ability of plants to absorb phosphate is one of determinants of plant growth and development.Phosphate mobilization into the plant is a complex process in which the absorption and translocation of this major nutrient are determined largely by the phosphate transporters.Plants regulate the expression of Pi transporter genes to adapt low available P environment by enhancing their ability to acquire and transfer Pi.One member of the Phtl family,the LePT1 gene,which was derived from the tomato,encodes a high-affinity phosphate transport protein.LePT1 is transferred into tobacco,a model plant,to study its adaptability to a low-phosphate environment.This research lays the foundation for the creation of new strain crops through transgenic technology that are resistant to the problems of a low-phosphate environment.A plant expression vector was constructed by inserting the phosphate transporter gene,LePT1,which was cloned from the tomato genome,into pCAMBIA2300.An agrobacterium-mediated system was used to transform tobacco and acquire transgenic plants.Analyses of the transgenic plants by PCR and RT-PCR indicated that the exogenous gene was integrated into and expressed by transgenic plants,conversion rate is 68%.The expression of LePT1 in tobacco was induced by low-Pi conditions,and when the gene was induced by low-Pi conditions for 12 hours,the expression of LePT1 significantly increased and then decreased soon after.In addition,LePTl was expressed in both the roots and leaves,but the expression level in roots was higher than in leaves.The growth characteristics of T1 generation transgenic plants were examined,transgenic tobacco plants plants growth was better than the wild-type plants.The Pi content were significantly different(P<0.05),where the Pi content of transgenic tobacco plants was 1.62 times higher than the wild-type plants.It showed that in the Pi-deficient conditions,transgenic plants were more effective than the wild-type plants.
