In this study,we studied the protective effect of BMSCs derived from bone marrow mesenchymal stem cells(BMSCs)on HT22 cells injured by H2O2.The primary bone marrow mesenchymal MSC cells were isolated from the bone marrow of SD rats.After the cells were passaged three times,the morphology was observed and the BMSC was treated with serum-free medium for 24 h.The supernatant of BMSC was collected firstly,then the Exosomes of BMSCs in rat bone marrow mesenchymal stem cells were collected by gradient centrifugation.Firstly,TEM was used to identify Exosomes from the size.The total protein of Exosomes was extracted and the specific antigen molecules were identified by immunohistochemistry.The model of H2O2 was established by HT22 cells and divided into blank group: normal growth of HT22 cells,control group: treated with 0.8 mM H2O2 for 2 hours,the Exosome + H2O2 group : Exosome and H2O2 were added at different times.The changes of cell morphology were observed by fluorescence staining.MTT assay and SOD and LDH were used to detect the toxic effects of different treatments on HT22 cells.After incubation for a period of time,the growth status was good.BMSC cells were long fusiform adherent,disordered and swollen.After centrifugation,the culture medium can be vesicles with round and complete cell membrane.This foam expresses exoskeletal specific membranes CD9,CD81 and HSP70 antigens.After the H2O2-induced HT22 cell skeleton changes,the cytoskeleton of HT22 cells treated with Exat-treated HT cells has been restored,sthe rat bone marrow mesenchymal stem cells Exosome can significantly improve the morphological damage of HT22 cells after oxidative damage MTT assay showed that the proliferation inhibition rate of HT22 cells in the injured group was increased,and the proliferation inhibition rate of the rat bone marrow mesenchymal stem cells was decreased,which was significantly different from that of the H2O2 control group.At the same time,the release of LDH and the activity of SOD were increased in HT22 cells after H2O2 injury,but after the addition of Exosome can significantly reduce the LDH release and SOD activity.Using PKH26 to secrete the body of the staining,and then track its positioning in the HT22 cells and found that Exosome cells were collected near the cells and around the collection.Finally,we found that Exosomes derived from rat bone marrow mesenchymal stem cells(BMSCs)have a protective effect on H2O2-induced injury of HT22 cells.The mechanism may be related to the enhancement of antioxidant capacity of HT22 cells.
