In recent decades,plant can protect themselves by the synthesis of trehalose to response to abiotic stress such as drought,cold.However,the mechanisms of trehalose metabolism protecting plant to response to abiotic stress are not clear.Trehalose synthesis is most important for organisms.Microarray data demonstrated that TPP4 gene expression will be increased obviously when the plant suffered adiotic stress.In order to explore the relationship between TPP4 gene and ABA signaling pathways,we ordered the mutant tpp4-1,tpp4-2,tpp6,akin10 from ABRC.We found that the mutant tpp4 was lack of sensitivity to ABA in terms of root tip growth.Compareqd with WT,the root growth of mutant tpp4 is faster than WT under 25 ?M and 50 ?M ABA treatment.In addition,the water loss rate of mutant tpp4-1,tpp4-2 significantly higher than that of WT in vitro.It was reported that the mutant tpp6 was lack of sensitivity to ABA,in terms of seed germination and stomatal aperture.Besides,T6 P inhibits Sn RK1 to activate biosynthetic processes in growing tissues of Arabidopsis thaliana.However,Sn RK1 can specifically phosphorylated AREBPs,the transcription factors in ABA signaling pathway.In order to explore the link between TPP4 gene and ABA signaling pathways,we gained the double mutant tpp4tpp6,tpp4akin10 by hybridization technique.We looked at the effect of T6 P in stomatal aperture and closure.When treated with 10 ?M T6 P,the stomatal closure of WT is obvious,but that of double mutant tpp4akin10 showed no significances.What's more,T6 P could not induce stomatal closure in the mutant ost1.T6 P can alter the expression quantity of AKIN10 and OST1 gene in flurescent quantity PCR experiments.The trend of gene expression is consistent with the results of stomatal experiments.We discovered that T6 P,the intermediate product of trehalose synthesis pathway,could indirectly affect the ways of stomatal movementwhich mediated by ABA signaling pathways through Sn RK1/Sn RK2.6 and the ABA signaling pathways could modulate on genes expression related to trehalose synthesis pathway.
