Most plants can not survive or produce biomass in high salt environments,while some plants can grow normally in environments with hypersaline habitat.Helianthus tuberosus is herb plant that can grow normally in the soils of high salinity.Because of its tuber rich in inulin and other ingredients with medicinal value,in agriculture and medicine and many other industries have higher application value.Therefor,Helianthus tuberosus become a hot research.The physiological mechanism of salt tolerance in Helianthus tuberosus has been studied.The transcriptional analysis of the root of Helianthus tuberosus under salt stress was conducted to facilitate the study of gene expression changes in roots of Helianthus tuberosus in response to salt stress from the perspective of whole genome.In our experiment Illumina Hiseq technology of NGS was used for transcriptional sequencing of root of Helianthus tuberosus for 150 mM NaCl dispose for 15 days and control group.The changes of genome of Helianthus tuberosus roots under salt stress were analyzed.And a lot of up-regulated and down-regulated genes were found under salt stress in Helianthus tuberosus.And there were 11 differentially expressed genes were screened under salt stress for real-time quantitative PCR to validate the data.The findings are as follows:(1)The overall sequencing output were over than 1.5billion and 1.7 billion clean reads of control and treatment of Helianthus tuberosus root in 50.25 G database and the clean reads exceed 94%.(2)190567 Unigenes was assembled from the clean read of tanscriptome.There are 65939,30259,21718,47089,46340,47066,21735,Unigenes were categorized into NR(NCBI nonredundant protein sequences),NT(NCBI non-redundant nucleotide sequences),KO(KEGG Ortholog database),SwissProt(A manually annotated and reviewed protein sequence database),PFAM(Protein family),GO(Gene Ontology)and KOG(Clusters of Orthologous Groups of proteins)databases respectively.However,80511 Unigenes were not categorized.(3)Expression pattern of the differentially expressed genes showed that,all the differentially expressed genes(DEGs)of Helianthus tuberosus roots in response to 150 mmol / L NaCl stress were annotated in the GO database,found that the component of metabolic process categorize 2315 DEGs.In the KEGG database,the most significant genes were included in the ribosome Ribosome pathway,in including 134 genes up-regulated and only 2 genes down-regulated.(4)11 differentially expressed genes were screened for real-time quantitative PCR.The resulting expression trend was consistent with the results of the transcriptome.
