In this paper, the accumulation, purification, antioxidant activity, lipid-lowering effect and anticoagulant activity of extracellular polyphenols from Lachnum singerianum(LSP) were studied. The main results were as follows:Saccharomyces cerevisiae polysaccharide as a fungal elicitor was added to fermentation broth of Lachnum singerianum and studied its effects on polyphenols accumulation. The result showed that the fungal elicitor could improve the activities of nitric oxide synthase and phenylalanine ammonia lyase and increase accumulation of biomass and extracellular polyphenols. Compared with the normal group, the accumulation of polyphenols in the 80 ?g/mL fungal elicitor group was increased by 28.0%, which showed that the fungal elicitor could increase extracellular polyphenols accumulation. LSP was separated and purified by Sephadex LH-20 chromatographic columns to obtain the main fraction LSP-1. According to the results of UV-Vis, FT-IR, and GC-MS analysis, LSP-1 contained phenolic acids and polyhydric phenols has been demonstrated.Antioxidant activity assay in vitro showed that, at a concentration of 2.5 mg/mL, LSP-1 has significant reducing power, and the scavenging effect on O2-and DPPH·were 54.5% and 70.4%, respectively. The lipid-lowering activity of LSP-1 has been researched and the result showed that LSP-1 could effectively reduce average weight gain, feed efficiency and liver index of high-fat-diet mice. Meanwhile, LSP-1 could lower triglyceride and total cholesterol levels in serum and liver. Similar results were observed for atherosclerosis index, low-density lipoprotein cholesterol levels, and activities of alanine aminotransferase and aspartate aminotransferase. But high density lipoprotein cholesterol was significantly increased. Further studies showed that these activities of SOD, GSH-Px, and CAT in serum and liver were significantly increased, but MDA content was decreased. Moreover compared with the high fat diet control group, swelling and fat vacuoles in liver were reduced in high dose group. The results suggested that LSP had hypolipidemic effect in high-fat-diet mice effectively.Anticoagulant activity study showed that LSP-1 could extend PT, ATPP, TT of plasma in vitro. Via the anticoagulant activity assay in vitro, LSP-1 could significantly prolong CT, BT, PT, ATPP, TT, reduce the content of FIB and enhance the AT-III activity of blood hypercoagulable state animal model induced by adrenaline. The results showed that LSP-1 had significantly anticoagulant effect.
