Elimination Of Allergens From Litopenaeus Vannamei By High-pressure Treatment Combined With Enzymolysis

To eliminate the allergens in Litopenaeus vannamei,three methods were separately used to deal with the protein of this shrimp,shrimp paste and the whole shrimp,that is high-pressure treatment,papain enzymolysis after high-pressure treatment and pressurization in the presence of papain.To the protein of this shrimp,the resulrs showed that the optimal condition for high-pressure treatment was performed under 150 MPa pressure for 35 min at 45℃,and the OD492 nm value of antibody reaction is 0.1977,the allergenicity was cut down 66.2% compared with the positive reaction.The optimal condition for papain enzymolysis after high-pressure treatment was performed under 350 MPa pressure for 20 min at 45℃,and the OD492 nm value of antibody reaction is 0.0492,the allergenicity was cut down 91.6% compared with the positive reaction.The optimal condition for pressurization in the presence of papain was performed under 300 MPa pressure for 35 min at 45℃,and the OD492 nm value of antibody reaction is 0.05,the allergenicity was cut down 91.5% compared with the positive reaction.The three methods was effective for allergen elimination,it was better that papain enzymolysis after high-pressure treatment and pressurization in the presence of papain.To the the shrimp paste,the results showed that the optimal condition for high-pressure treatment was performed under 200 MPa pressure for 35 min at 40℃,and the OD492 nm value of antibody reaction is 0.1986,the allergenicity was cut down 66.1% compared with the positive reaction.The optimal condition for papain enzymolysis after high-pressure treatment was determined as follows: substrate concentration is 10 g/100 mL,hydrolysis temperature is 60 ℃,enzyme/substrate mass ratio is 1:200,and the OD492 nm value of antibody reaction is 0.0487,the allergenicity was cut down 91.7% compared with the positive reaction.The optimal condition for high-pressure treatment in the presence of papain was performed under 300 MPa pressure for 35 min at 40℃,and the OD492 nm value of antibody reaction is 0.0516,the allergenicity was cut down 91.2% compared with the positive reaction.The three methods is effective for allergen elimination,but it was better that papain enzymolysis after high-pressure treatment or high-pressure treatment in the presence of papain.Two methods were used to deal with the whole shrimp,that is high-pressure treatment and high-pressure treatment in the presence of papain.The results showed two methods are effective for allergen elimination,but it was better that high-pressure treatment in the presence of papain.The optimal condition for high-pressure treatment was performed under 100 MPa pressure for 15 min at 25℃,and the OD492 nm value of antibody reaction is 0.1925,the allergenicity was cut down 67.1% compared with the positive reaction.The optimal condition for pressurization in the presence of papain was performed under 450 MPa pressure for 55 min at 40℃,the ratio of enzyme to substrate rate is 1:130,and the OD492 nm value of antibody reaction is 0.084,the allergenicity was cut down 85.6% compared with the positive reaction.At the same time,allergen protein can release from whole shrimp during the two methods.When the method of high-pressure treatment in the presence of papain was used to deal with the whole shrimp,the concentration of saline water had a certain impact on the eliminating effects of allergens and the dissolution of allergen protein.the conditions of the whole shrimp was dealed by high-pressure treatment in the presence of papain as follows: the concentration of saline water is 1.0%,pressure is 450 MPa,temperature is 40℃,dwell time is 55 min,the ratio of enzyme to substrate rate is 1:130,and the OD492 nm value of antibody reaction is 0.074,the allergenicity was cut down 87.4% compared with the positive reaction.
