Dencichine is the hemostatic ingredient in traditional Chinese medicines Panax notoginseng, the hemostatic activity of dencichine is better than tranexamic acid, and it is a good hemostatic agent. Gastrodin is the main active ingredient in Gastrodia elata Blume with analgesic, sedative, hypnotic, anti convulsion, protection of the nervous system and anti-aging and other physiological functions. Dencichine and gastrodin are water soluble compounds and they are highly polar and small molecular compounds, which are very difficult to isolate and purify. The chemical constituents of Panax notoginseng and Gastrodia elata Blume are complicated and the traditional separation methods are inefficient. Therefore, it is necessary to establish an efficient method to separate and purify dencichine and gastrodin.In this research, we took full advantages of the specific recognition on molecularly imprinted polymers and prepared polymers with highly selective recognition and high adsorption capacity on dencichine and gastrodin by molecularly imprinting technique. Finally, the polymers were used as solid-phase extraction(SPE) adsorption material to separation of dencichine and gastrodin form Panax notoginseng and Gastrodia elata Blume.1. Selective preparation of dencichine by Surface molecularly imprinted polymersSDMIPs(surface dummy molecularly imprinted polymers) were prepared by using silica gel as the carrier, acrylamide(AM) as functional monomer, ethylene glycol dimethacrylate(EGDMA) as crosslinking agent and acetonitrile as porogen. The binding experiment revealed that the polymers with D-leucyl glycine(LG) as template molecule showed the best adsorption properties. The maximum adsorption capacity of polymers were 0.185 mmol/g and it only took 15 min to achieve adsorption equilibrium. The purity of dencichine was 98.7% and the average recovery rate was 83.7% when the polymers were used as SPE adsorption material to separation of dencichine. The reserach showed that the surface molecularly imprinted polymers were suitable for SPE adsorption materials to separation and enrichment of dencichine.2. Selective preparation of gastrodin by hydrophilic molecularly imprinted polymersOn the basis of previous work of our group, we prepared H-MIPs(hydrophilic molecularly imprinted polymers) by hydrolysis of ester groups on the surface of hydrophobic molecularly imprinted polymers of gastrodin. The FT-IR showed that part of the ester groups on the surface of polymers turned into hydroxyl groups and the hydrophilic test results showed that the polymers had good hydrophilicity. The binding experiment revealed that compared with the hydrophobic molecularly imprinted polymers, H-MIPs had a faster adsorption rate and higher adsorption capacity on gastrodin. The purity of gastrodin was 98.8% and the average recovery rate was 84.9% when H-MIPs were used as SPE adsorption material to separation of gastrodin. The reserach showed that H-MIPs were more suitable for SPE absorbing materials for the selective separation of gastrodin.
