| Mycotoxins are toxic secondary metabolites produced by fungi.The grain may be infected by mycotoxins during their growth,harvest,storage,processing and transpotation.Among the mycotoxins identified to date,the mycotoxins which are related with the health of people contain aflatoxins,deoxynivalenol,zearalenone,and so on.In order to controlling the contamination of mycotoxins,this thesis studied and detected the aflatoxin B1,deoxynivalenol and zearalenone in different kinds of grain.The main conclusions were as follows:(1)Detecting the aflatoxin B1,deoxynivalenol and zearalenone in different kinds of grains by multifunctional column clean-up and high performance liquid chromatography(HPLC).It shows that the aflatoxin B1 was widely found in different kinds of grains.Corn,wheat and peanut were infected by aflatoxin B1 seriously,the detection rate both are above 80%.Besides,the contents of deoxynivalenol found in corn,wheat and oats were higher than other kinds of grains.Zearalenone exists mainly in corn.Deoxynivalenol content is generally low in freshly harvested corn,but after storage time,the content of deoxynivalenol increased.This shows that when the environmental conditions are suitable,deoxynivalenol will grow.(2)Through the simulations of high temperature and humidity,low temperature and enclose,ventilation and cool,using established analytical methods,this thesis investigated that the storage conditions were very important to the mycotoxins in corn.AFB1 and DON increase most in storage in high temperature and humidity.So,this condition is not suitable for corn.The best way to storage the corn is under ventilation and cool condition.(3)The most stable results were obtained when corn samples were extracted with 70% acetonitrile using phosphate buffer solution with 0.25% gelatin as the masking agent.The findings of this study can provide a reference for the development of highly stable ELISA kit for the detection of deoxynivalenol in corn. |
