| Dictyophora belongs to Gasteromycetes,Phallales,Phallaceae,Phallus indusiatus.Dictyophora has many functions,such as nourishing function,tonifying qi,boosting brain,relieving uneasyness of mind,building fitness,boosting qi and nourishing yin,moistening lung,reducing cough,clearing heat and promoting diuresis.It is proven that Dictyophora both has good antineoplastic activity and special antiseptic.However,the research on chemical components and pharmacological function was still ignored.Until recent years,with the development of modern analytical technology and new technology of food industry,more attentions were paid to chemical components,pharmacological values,functional study and new product exploitation.Extraction condition,antibacterial properties from antibacterial substance of Dictyophora and ethyl acetate extract were analyzed,to prove that the extraction has the antibacterial properties.It aslo has a good industrial prospect for the development of functional food of fungi and production of extract from Dictyophora.(1)Regression equation was set up by quadratic orthogonal rotating design,which was based on three factors containing solid-liquid ratio,temperature,time.The model could accurately predict the extraction effect of antibacterial substance within the scope of the trial.The results of the influence of extract effect were as follows:temperature>time>solid-liquid ratio.The optimal extraction conditions were solid-liquid ratio of 1:25,temperature of 75℃,time of 3h.(2)Bacteriostatic experiments of four strains containing Escherichia coli,Salmonella,Staphylococcus aureus,and Bacillus subtilis were carried out by continuous dilution.The result indicated that ethyl acetate extract had a more obvious inhibitory effect on Escherichia coli and Salmonella than that of Bacillus subtilis and Staphylococcus aureus.(3)Inhibitory effects on the growth of Escherichia coli and Staphylococcus aureus were more obvious than that of Bacillus subtilis,Salmonella and Pseudomonas when the extract was treated with 121℃.However,inhibitory effects on the growth of Pseudomonas,Salmonella,Staphylococcus aureus and Bacillus subtilis were more obvious than that of Escherichia coli,when the temperature was 105℃.(4)Ethyl acetate extract had certain antibacterial activities after treating with 105℃ for 15min,it was indicated that they had certain heating stability.However,after treating with 121℃ for 15min,some lost partial sterilization.The results were as follows:Ethyl acetate extract had inhibitory effects on the growth of Pseudomonas,Staphylococcus aureus,Escherichia coli,Salmonella enteritidis,Bacillus subtilis and Escherichia coli after treating with 105℃ for 15min.And Ethyl acetate extract had inhibitory effects on Staphylococcus aureus and Escherichia coli,but hadn’t effects on Salmonella enteritidis,Bacillus subtilis and Pseudomonas.It is necessary to avoid excessive temperature when we process food.Because it might lead to the loss of nutritional ingredients and do harm to the antibacterial substance.(5)Inhibitory effects on the growth of strains after treating with 105℃ for 15min were obvious than that of 121℃ for 15min.So it was necessary to avoid the processing temperature.(6)The scope of pH value of the ethyl acetate extract treated with 105℃ for 15min and 121℃ for 15min was 6 to 8.And the blank control group had antibacterial effects when the pH value was 4,and had not antibacterial effects on any other pH value.Maybe the strains have the optimum growing environment with the neutral condition,instead of the acidic condition.(7)Bacillus subtilis treated by extracting Dictyophora and blank group were researched by macro protein SDS-polypropylene amide gel electrophoresis.The result showed that there is a difference containing 18.4kDa between experimental groups and blank groups.Maybe Dictyophora extract inhibit bacterial protein expression,and it can be concluded that Dictyophora extraction has a certain inhibitory activity. |
