Porphyrin and its derivatives were widely found in nature,and plays an important role in the life activities.Modern research showed that porphyrins had an important position in biological medicine,materials,catalysis and actinic areas.As an important photosensitizer,porphyrin is an important link in photodynamic therapy.Now a lot of article on porphyrin as a photosensitizer applied photodynamic therapy literature had been consulted and briefly described the progress of porphyrin chemistry and researched progress in the field of anti-tumor.In this thesis,five conjugated porphyrin compound were designed and synthesized,the target compounds and intermediates were characterized by UV,IR,NMR,MS and other means.The interaction of five porphyrins with calf thymus DNA was studied by ultraviolet spectrum and fluorescence spectra.At the same time,with the aid of circular dichroism,the binding modes were the outside binding and outside stacking for Por1 in high ratio of DNA base pairs(r=0.05,r=[Porphyrin]/[DNA]),while Por2 showed the intercalative interaction and outside stacking mode in the same condition.DNA could be inserted with the DNA phosphate backbone phase by electrostatic action by Por3,and it could itself be deposited on the surface of DNA.The binding modes were the intercalative interaction and outside binding for Por4 and Por5.At the same time,measuring their singlet oxygen quantum yield by indirect assay.It is a high yield of singlet oxygen for metal porphyrins(Por1,Por2),while water-soluble porphyrin(Por3)also showed good singlet oxygen yield.Morphological images of ct-DNA-porphyrins complex,both shape and size,were observed by AFM.After Por2 and Por3 were photocleaed to ct-DNA in the presence of light irradiation,the shape and size of DNA blank,ct-DNA-porphyrin in absence of light irradiation changed.It further proved that the influence of shape and size of biomolecules and important.So conjugated porphyrin were potential photosensitizer drugs.It was laied the foundation for cell research in vitro.
