| Walnuts are rich in protein,vitamins,minerals and polyphenol,has high nutritional value.In industrial production,the by-product of Walnut kernel pellicle contains most polyphenols of active ingredients with antioxidant,antitumor,antibacterial and anti-inflammatory properties.Walnut Peel is widely used traditional method of NaOH,and lots of research on walnut kernel pellicle is just the study of fresh walnut kernel pellicle.Therefore,subject to the walnut in fen yang of Shanxi province as raw materials,research on the effect of a new method of peeling walnut kernel pellicle which is salt solution dipping method.Meanwhile,collecting by-products of walnut kernel pellicle and extracting the polyphenols.And then do a preliminary purification,identification and antioxidant activity for walnut kernel pellicle.Main results are as follows:(1)Removal of Walnut seed coat using a new method of salt solution dipping method,than NaOH method,Walnut nutrition preserved,without limiting amino acids and amino acid score is higher than 0.26,not saturated fatty acid reduction is the latter half of polyphenols in the walnut seed coat are 15.32%and 4.26%respectively.Shows that salt solution dipping method will work better,and be able to keep more of the active ingredient,and increase added value.(2)Ultrasound-assisted extraction of walnut kernel pellicle polyphenols,ultrasonic power 300W,frequency 50Hz,extracted solid-liquid ratio is 1:50,extraction temperature is 50 degrees,time to 50min,70%ethanol,in the conditions of Walnut seed polyphenols extraction rate of 15.34%.(3)The comprehensive analysis of adsorption effects of macroporous adsorption resin NKA-9 the best,after the analysis of dynamic,Walnut seed polyphenols purity is 82.14%.(4)Color reaction showed that Walnut seed polyphenolic substances containing polyphenols and phenolic hydroxyl groups of characteristic reactions.UV-visible absorption spectra shows that extracts containing polyphenols and flavonoid compounds.Infrared analysis results showed that Walnut seed polyphenols contained in acetylation acid polyphenols;Walnut kernel pellicle HPLC chromatogram of polyphenol that Walnut seed polyphenols may contain a main material for Gallic acid,catechin,chlorogenic acid,epicatechin,caffeic acid,vanillic acid,syringic acid,quercetin,molar ratio of 19.192:30.836:9.341:13.936:0.563:7.824:0.651:5.406.(5)The results of research of walnut kernel pellicle polyphenols in vitro antioxidant activity showed that Walnut seed polyphenols has strong restoring force,hydrogen peroxide(H2O2),DPPH-free radical scavenging,hydroxyl radicals(·OH)and lipid peroxides.In a certain range,the antioxidant capacity increases with the concentration,and similar with tea polyphenols,slightly less than the VC.(6)Selecting the D-Galactose aging mice,after seven weeks,the results of the Morris water maze test in mice show that molds success,and the memory ability of dose group mice was significantly stronger than model mice.Walnut kernel pellicle polyphenols has some memory effect,and is same to the effect of tea polyphenols.(7)Selecting the D-Galactose aging mice,and gavaging the mice in 50,100,200mg/kg · bw · d dose of walnut kernel pellicle polyphenols in aqueous solution.While gavaging in tea polyphenols as a control.The results showed that Walnut seed polyphenols may be able to inhibit fat formation,reduced brain cell apoptosis rate and active oxygen in brain and liver cells levels,reduce the oxidation of unsaturated fatty acids protect brain cells from damage.Also may be able to effectively improve brain tissue,serum and T-AOC,CAT and SOD vitality in the liver,lower MDA content in brain tissue,serum and liver,while improving GSH-PX activity in the brain and liver.Walnut seed polyphenol antioxidant effects of a dose-dependent,and is same to the effect of tea polyphenols. |
