| Based on the indoor cultured cyanobacteria and the wild algae as the research object,the concentration of the extracted phycocyanin as an index,by means of fluorescent assay to detect concentration of phycocyanin,based on the crushing methods and the selection of extraction agent,respectively explore the relation between algae biomass and concentration of phycocyanin in the water /sediment samples.The experimental results show that:In water samples,use the microscope to count the biomass of cyanobacteria and the extraction efficiency of phycocyanin under the same experimental conditions,results show that: the lyophilization on algae cells crushing efficiency is higher than repeated freezing and thawing method.When the cells crushing method and other experimental conditions keep invariant,compared with deionized water,phosphate solution and Tris reagent of algal blue protein found that the extraction efficiency of deionized water to the algae blue protein extraction efficiency is highest.In repeated freezing and thawing experiment,freezing temperature and extraction agent adding order,had no significant effect on algae cell breakage rate.In freezedrying experiments,cyanobacteria vacuum suction filter on the membrane filter can improve the breakage rate of algal blue protein.Using fluorescence assay to detect the phycocyanin.Efficient pre-treatment technology for environmental monitoring in lakes can’t only achieve a rapid and accurately quantitative algae biomass method,but also can prevent and control of cyanobacteria blooms.Detecting sediment samples found that under the same experimental conditions,freeze-dried crushing method and repeated freezing and thawing method have the same efficiency for the algae cells in sediments.The sediment moisture content had a great influence on the result of the experiment,because of adsorption and masking effect.The higher the moisture content of sediment,extraction efficiency of phycocyanin is higher.Hence,improve the moisture content of sediment is one of the most important measures to improve the certainty.Algae biomass and concentration of phycocyanin in the sediment has a good linear correlation,therefore can through the concentration of phycocyanin to calculate the algae biomass.Contrast microscope counting method,flow cytometry method and phycocyanin indirect quantitative methods,the microscope and flow cytometry instrument can accurately count of algae biomass of water,the sediment of blue-green algae to do.And indirectly by algae blue protein quantitative algae cells can not only accurately calculating the biomass of algae in water,also can accurately calculate the biomass of cyanobacteria in sediments.And the experiment results have good stability and high accuracy. |
