In order to develop the potentiality of Talaromyces wortmannii LGT-4 on secondary metabolism, we have taken the research on fungal culture medium, and the rice medium and zeistic medium were choosen to be the fermentation cultures of T.wortmannii LGT-4. Eleven compounds were isolated by the morden chromatographic techniques, including silica gel column chromatography, TLC, HPLC. All of the compounds were elucidated by the 1D and 2D NMR spectroscopy, ESI-MS, mosher’s method,single-crystal X-ray crystallography and comparing the experimental and calculated electronic circular dichroism(ECD) spectra. Six of the compounds were proved to be new compounds. All of the compounds were evaluated for their bioacitivities including Monoamine oxidase inhibitory activity, acetylcholinesterase inhibitory activity and cytotoxic activity. The main result were shown as the following:(1) We compared the chemical composition of the secondary metabolites of T.wortmannii LGT-4 cultured on twelve kinds of culture medium by HPLC technique.The rice medium and zeistic medium were chosed to continue our study on Chemical Constituents of T. wortmannii LGT-4.(2) Three compounds were isolated from the extract of the fungus which was cultured on the rice solid medium. And, two compounds were identified as the new compounds, named deacetylisowortmin A(1) and deacetylisowortmin B(2),seperatively. The left one was a known compound named5-hydroxy-2-(2-hydroxy-6-methoxy-4-methylbenzoyl)-3-methoxybenzoate(3)..(3) Eight compounds were isolated from the extract of the fungus which was cultured on the zeistic solid medium. Four compounds were identified as known compounds named entonaemin A(8), meso-1,4-Bis(4-methoxyphenyl)-2,3-butanediol(9), monomethylsulochrin(10) and(4S,4a S,5R,6a S,9a R)-5-(acetyloxy)-1-(aminomethylene)-4,4a,5,6,6a,8,9,9a-octahydro-11-hydroxy-4-(methoxy methy)-4a,6a-dimethylcyclopenta[5,6]naphthol[1,2-c]pyran-2,7,10(1H)-trione(11). And the others were identified as new compounds named wortmannines A-D(4-7). Among these new compounds, wortmannines A-C(4-6) were proved to have novel carbon skeletons.Wortmannine A(4) was a dinor-carbon wortmannin deravative with an unusual five-membered B ring, wortmannine B(5) was a wortmannin deravative bearing an unusual five-membered B ring and lactam E-ring, and wortmannine C(6) was a wortmannin deravative bearing an unusual five-membered B ring and a lactone E-ring.(4) All of the compounds were evaluated for their monoamine oxidase inhibitory activities, acetylcholinesterase inhibitory activities and the cytotoxic activities. Only entonaemin A(8) showed anti-HL60 activity with IC50 s values of 15.0 μM(treated for24h) and 12.4 μM(treated for 48h).
