The papermaking wastewater from a Papermaking Plant in Shandong,with COD of about 12000 mg/L and pH 12-13,is characterized by non-degradable organic pollutant and complex water quality.The samples were collected from pulp mill wastewater sediment.By the methanogenic community enrichment and the clone library construction of the McrA gene to analyzed the composition of methanogens.On this basis,build acid-producing and methane-producing mixed strains by using some acid producing strains screening from papermarking waste water and methanogenesis bacteria liquid enrichment from papermarking waste water sediment.(UASB)was used to domesticate the papermarking waste water and DGGE was used to analysis the role of the added microbial community.Second,the stain named K.oxytoca which can produce PD from glycerol was isolated from the methanogenesis bacteria liquid.At the last,the PD fermentation conditions from glycerol by K.oxytoca KL1001 strain were investigated.1.We obtained the sodium formate and H2/CO2 enrichment of methanogenesis bacteria liquid respectively by using the methanogens enrichment medium to enrichment he methanogens from the samples collected from papermaking waste waterr sediment.We analyzed the two methanogenesis bacteria liquid by building a genomic library of the McrA gene and found a lot of Methanoculleus sp.2.We builded the acid-producing and methane-producing mixed strains by using some acid producing strains(Clostridium sp.YC3,Halomonas sp.Y2 and Halomonas sp.19-A)screening from papermarking waste water and methanogenesis bacteria liquid enrichment from papermarking waste water sediment.Through domestication,we implement the preliminary processing of the strong alkaline paper making black liquor.Analyzing the roll of produce acid bacteria and methane-producing microbes added in the reactor by using DGGE,we found that the content of the adding Clostridium sp.was increased with the increasing of water sewage concentration while the content of the adding Halomonas sp.was decreased with the increasing of water sewage concentration but could maintain a certain amount.The dominant Methanoculleus sp.in the methanogenesis bacteria liquid was no found before the addition of methane-producing mixed strains,but showed stable existence after addition.After all,the acid-producing and methane-producing mixed strains in the UASB reactor can stable exist and play an important role.3.At the last,the stain named K.oxytoca which can produce PD from glycerol was isolated from the methanogenesis bacteria liquid.The PD fermentation conditions from glycerol by K.oxytoca KL1001 strain were investigated.Micro-aerobic was suitable for PD fermentation by strain KL1001.In the co-fermentation,the sucrose added in the medium accelerated the production of PD.PD production was enhanced by fed batch fermentation.
