As one of main active components of the Epimedium,polysaccharide is paid more and more attention increasingly for its good function of enhancing immunity,stimulating platelet production and anti-tumor activity.In this thesis,the determination and preparation process of Epimedium polysaccharide were investigated.Main research contents and innovations:The phenol-sulfuric acid method was widely applied in the determination of polysaccharide.There were many improved phenol-sulfuric acid methods which had been researched.On this basis,we did further study to set up a new method named Improved-Differential phenol-sulfuric acid method which was simple,accurate,suitable for the determination of polysaccharide.The solving measures of membrane fouling were investigated which as the bottleneck limiting the wider application of the membrane separation technology in purification of traditional Chinese medicine.Close to the room temperature operation for thermal components,and the lack of obvious disadvantages such as phase change.Therefore,application of this technology in purification of Epimedium polysaccharide was studied in this experiment.The bottleneck issue were explored in this experiment,study on the effect of membrane fouling with different pre-treat methods,and a satisfactory result has been obtained.The utilization of medicinal herbs,and increase the availability of medicinal materials and profits,decrease production costs,while also reducing waste and protecting the environmental,comprehensive utilization technology of the active ingredient in Epimedium was explored.In this thesis,papain was first used as deproteinization method to remove protein from Epimedium polysaccharide which prepared by membrane separation technology.Influencing factors of enzyme method on the deproteinization were investigated,and compared with Sevag method.The results showed that the method of enzyme-sevag was effective method to remove protein from Epimedium polysaccharides.At last,the different extraction methods of Epimedium polysaccharides were researched.The extraction duration,temperature and solid-liquid ratio of water extraction method and Ultrasonic extraction method were studied,and used orthogonal experimental design to optimum conditions.Above research is benefit for the development and utilization of Epimedium herbs,which will be helpful for similar work and projects.
