Lonicera edulis are much richer in anthocyanin content than other fruits and vegetables and widely grown in Changbai Mountain with low price,but the extent of its comprehensive utilization are lower.Anthocyaninin Lonicera edulis is safe,nontoxic,rich in resources and has certain nutritional and pharmacological effects.Lonicera edulis Anthocyanin has been widely used in cosmetics,food,medicine industry for its great development potential.In this study,Ultrasonic-microwave was applied to extract the anthocyanins of Lonicera edulis.The anthocyanins was purified with macroporous adsorption resin and the antioxidant activity was analyzed and microcapsules was prepared,with a view to develop a new way of efficient extraction of anthocyanins and providing a theoretical basis for the comprehensive utilization of Lonicera edulis.1.The method of traditional solvent extraction and pulsed electric field and Ultrasonic-microwave assisted extraction were used to extract anthocyanins from Lonicera edulis.The optimized extraction conditions and anthocyanin content were compared.The results are as follows:(1)The contrast test shows that anthocyanins extraction yield by Ultrasonic-microwave are higher,less consuming.(2)Response surface was applied to get the quadratic regression equation between anthocyanins extraction yield(Y)from Lonicera edulis.by Ultrasonic-microwave and ethanol volume fraction(X1),treatment time(X2),microwave power(X3),solid-liquid ratio(X4):Y=-105.46+2.08X1+21.47X2+1.22X4-0.10X1X2-0.10X2X4-0.01X12-1.49X22we obtained optimum parameters,which were ethanol volume fraction 59%,treatment time 3 min,microwave power 287(W),and the ratio of material to liquid1:60(g/m L),the maximum amount of anthocyanins was 33.62 mg/g.2.Anthocyanins from Lonicera edulis were separated and purificated with six types of macroporous adsorption resins with different chemical and physical properties(HPD-700,HPD-100,HPD-80,NKA-9,D101,AB-8).The results are as follows:(1)Static absorption/desorption experiments showed that AB-8 resin had higher adsorption capacity and desorption ratio than the others.The time of adsorption toreach dynamic equilibrium was 5h.(2)The results of adsorption and desorption experiments showed that the p H3.0 of Lonicera edulis Anthocyanins has better absorption result,and the desorption capacity of 60% ethanol solution with p H 3.0 has better desorption result.(3)The results of the dynamic adsorption experiment showed that The extract diluted to 0.8mg /m L of 1.0m L/min flow rate can better finish the saturated adsorption.Anthocyanins started to break through with 175 m L of extract loaded and the complete desorption was achieved using 100 m L 60% ethanol with 1.0m L/min.(4)Lonicera edulis Anthocyanin of purification with AB-8 macroporous resin was purple black powder,the anthocyanin color value were 35.26 and 7 times higher than crude products,the recovery rate reached 89.53%.3.The antioxidant activities of anthocayanin from Lonicera edulis were evaluated by ability of free radical scavenging on DPPH,hydroxyl and superoxide anion,the inhibiting lipid peroxidation and the total reducing power.The results are as follows:anthocayanin from Lonicera edulis have shown antioxidant capacity in various systems in a certain concentration rage.With anthocayanin concentration increased,antioxidant capacity gradually increased.anthocayanin had a certain degree of antioxidant activity compared with the control Vc.4.Microencapsulation of anthocayanin from Lonicera edulis were prepared by using the method of vacuum spray drying.The results are as follows:(1)we obtained optimum parameters,which were the ratio of wall material to core material 6: 1,ratio of maltodextrin to acacia 3: 1,solid content 20%,inlet temperature 160℃,the maximum embedding rate was 96.85%.(2)The storage time of the encapsulated anthocyanins was improved.Innovation:Ultrasonic-microwave assisted Extraction has been used to extraction of nthocyanin from Lonicera edulis of Changbai Mountain for the first time,which explored a new way for efficient extraction of anthocyanin with higher yield and less solvent-consuming and relatively lower feed liquid ratio.
