| Ginger is one of the most characteristic traditional resources in our country,which plays an important role in the international market.Though ginger has excellent quality with a long history of production,the study level of its health function is relatively low.Most ginger is used as raw materials in the international market competition,and it seriously restricts the development of ginger health value.Therefore,developing the health function of ginger components has important significance for improving the whole study level in China.With high quality Laiwu ginger as raw material,gingerol and curcumin were extracted by simultaneous extraction while polysaccharide was extracted by compound enzymatic extraction.D-galactose and aluminium trichloride were used to construct mouse models of senile dementia.The mice were fed with gingerol,curcumin and polysaccharide extracted from ginger.And then the biochemical indexes of the mice’ various tissues were measured,followed by the significance of difference analysis,to determine anti-dementia effects of three components in ginger.The main results were as follows:(1)The extraction and purification of ginger polysaccharideThe optimized conditions of ginger polysaccharide by compound enzymatic extraction were: The best dosages of peak of alpha amylase,cellulase,pectinase and papain 2.5%,2.5%,1%,2%,pH value 5.2,the extraction temperature 55℃,and the extraction time 60 min.Under these conditions,the extraction yield of ginger polysaccharide was 22.18%.In the experiments of deproteinization from ginger polysaccharide,the optimized p H value was 3.5 by hydrochloric acid while the optimized pH value was 3 by trichloroacetic acid.And the optimized separation times were 5 by Sevage.Under these conditions,the deproteinization rates were 69.14%,60.82% and 62.14% respectively.In addition,by S-8 macroporous resin,the optimized mass of static adsorption was 4 g and the equilibrium time was 300 min.In the dynamic adsorption experiment,the optimized conditions were: the application amount 50 mL,application speed 1.5 mL/min,the elution speed 2.0 mL/min,the elution buffer dosage 3 times of the column volume.Comparing with other three methods,macroporous resin was the best to remove protein from ginger polysaccharide.The protein removal yield was 82.45% and polysaccharide loss yield was 10.53%.With the final concentration of ethanol-water solution being 50%,75% and 90%,the ginger polysaccharide was separated into three parts,named GP1,GP2,GP3.The yield of GP1 was 19.32%,GP2 was 69.72% and GP3 was 10.96%.Identified by SePhadex G-100 column,GP1 had two polysaccharide components,GP2 had a polysaccharide component combined with protein and GP3 probably had three small molecule polysaccharide components with a few of protein.(2)The study on anti-dementia effects of three ginger components on mouse models of senile dementiaThere were very significant anti-dementia effects of three ginger components.After high-dose curcumin was given to the mouse models of senile dementia by gavage,it increased the activity of SOD,CAT and GSH-Px and decreased the content of MDA in brains of mice both very significantly.Compared with the mouse models of administration,the effects reached up to 90.81%、92.63%、84.30% and 98.15% respectively.Besides,high-dose curcumin had significant effects on increasing the activity of SOD,CAT and GSH-Px and decreasing the content of MDA in liver,heart and kidney of the mouse models of senile dementia;After high-dose gingerol was given to the mouse models of senile dementia by gavage,it very significantly increased the activity of CAT and GSH-Px in brains of mice.Compared with the mouse models of administration,the effects reached up to 86.92% and 91.64% respectively.Besides,the body weight of the mouse models given high-dose gingerol by gavage very significantly increased by 67.07%;After high-dose polysaccharide was given to the mouse models of senile dementia by gavage,it increased the activity of SOD,CAT and GSH-Px and decreased the content of MDA in brains of mice both very significantly.Compared with the mouse models of administration,the effects reached up to 91.53% and 97.02% respectively.Among three ginger components,high-dose curcumin is the most effective one for anti-dementia. |
