| IntroductionOutlines the nuts contain nutrients and present research situation of nuts,intracellular thiol compounds is introduced in the important role of organisms and the analysis of the commonly used detection methods,and put forward such as glutathione in the nuts contain cysteine residues of sulfhydryl compounds determination method and the research significance,summarizes the topic of this research train of thought and main research content.Dertermination of Reduced Glutathione(GSH)and Thiols in Proteins(P-SH)in Nuts by High Performance Liquid Chromatography(HPLC)The contents of reduced glutathione(GSH)and protein thiols in prunus duleis,cashew nut,pistachio nut,macadamia nut were detected by pre-column derivatization HPLC method using 5,5-dithio-bis-2-nitrobenzoic acid(DTNB)as derivatizing agent with concomitant cell rupture sample pretreatments.The results showed that detected nuts contained relatively abundant GSH and thiols in P-SH.The amounts detected are:GSH=1.2303 μmol/g,P-SH=3.5414 μmol/g for pistachio nut,GSH=0.3069 μmol/g,P-SH=1.9001 μmol/g for prenus duleis,GSH=1.5708 p,mol/g,P-SH=0.5905 μpmol/g for macadamia nut,GSH=0.3373 μmol/g,P-SH= 2.2458 μmol/g for cashew nut respectively.The recoveries of the methods are 99.37%-101.45%,and detection limits are 0.1386 μmol/g for GSH and 0.2184 μmol/g for protein thiols.The results provided a useful data for the application of these nuts for the healthcare.Glutathione and Total Thiols are determined by spectrometry in NutsReduced glutathione(GSH)and total thiol in Nuts were determined by fluorophotometry and UV-vis spectrophotometry using O-phthaldehde(OPA)and 5,5’-dithio-bis-nitrobenzoic acid(DTNB)as derivatization agents.The results showed that detected nuts contained relatively abundant GSH and thiols in P-SH.The amounts detected are:GSH=1.0847 μmol/g,P-SH=5.0001 p,mol/g for pistachio nut,GSH=0.2890 μmol/g,P-SH=2.7627 μmol/g for prenus duleis,GSH=1.4720 μmol/g,P-SH=2.4774 μmol/g for macadamia nut,GSH=0.3255 μmol/g,P-SH=2.9302 μmol/g for cashew nut respectively.The recoveries for fluorophotometry and UV-vis spectrophotometry were 99.72%~100.90%and 98.66%~100.75%.Nuts may coexist in component’s influence on the determinationExplores the nuts of the vitamins,amino acids,unsaturated fatty acids and other active nutrients in this research reduced glutathione(GSH)and the influence of the determination of total sulphur(-SH).In spectrometry determination results show that high levels of vitamins and unsaturated fatty acids may produce certain effect,the results lead to the total sulphur content determination results on the high side,and the chromatographic method to determine the interfering substance to add of glutathione and had no effect on the determination of total sulphur content of the basic. |
