Capillary Electrophoresis?CE?is a kind of efficient and rapid new liquid separation technology.It is widely used in many fields such as life medicine,molecular biology,environmental science and food and drug quality monitoring,which provides a new feasibility for the detection of all kinds of substances to be tested.Sulfonamides are commonly used in the treatment of animal diseases.Estrogens are widely used in animal husbandry for fattening,especially in the dairy industry.But the pharmacological studies show that its unreasonable use of human endocrine interference,there is risk of teratogenic carcinogenesis.In order to ensure food safety and human health,it is necessary to test the content of sulfonamides and estrogens in animal food,and it is important to develop some high sensitivity,low cost,easy operation and popularize detection methods and techniques.In this paper,carbon fiber epoxy resin composite electrode was successfully used in electrochemical sensor,and it was applied to capillary electrophoresis detection.This article consists of the following four parts:Chapter1:IntroductionThis chapter mainly introduced the general situation of capillary electrophoresis,the pre-enrichment technology of the sample of capillary electrophoresis,summarized the influence of estrogen drug residues on human and environment and its research progress,and expounded the significance of this paper.Chapter 2:Preparation of micro-carbon fiber epoxy resin composite electrode for capillary electrophoresisDeveloped a simple method for making carbon fiber epoxy resin composite electrode for CE working electrodes.The composite electrode showed the characteristics of low noise level and strong stain resistance by comparison with the electrochemical characteristics of the pencil core epoxy resin composite electrode,ordinary disc electrode and microporous electrode.The separation and detection of the actual sample of sulfonamides by capillary electrophoresis can fully confirm the new detection electrode performance and advantages.The separation and detection of sulfonamides in the actual sample by capillary electrophoresis can fully confirm the performance,feasibility and analytical advantages of the new detection electrode.Simultaneously,because of its simplicity in design and manufacture,low cost,high success rate,it showed great potential in the field of analysis and application.Chapter 3:Determination of sulfonamides by micro-carbon fiber epoxy resin composite electrode for capillary electrophoresis.The carbon fiber epoxy resin composite electrode prepared as described above was uesd as a working electrode for detecting the residue of sulfonamides in the environmental water sample.The SAs of SM2,STZ,SMM,SD and SMZ were separated and detected.In the experiment,the sample was easy to handle,the instrument was simple,high sensitivity and good reproducibility.It had a certain reference value for the residual SAs in complex samples such as food.Chapter 4:Determination of the estradiol of estradiol in milk based on CZE-AD technologyA method for the accurate and rapid determination of estrogenic contaminants by capillary electrophoresis-amperometric detection was established.The composition of the background buffer system,the composition of the sample solvent,the injection time and the separation voltage were investigated.The three target components HEX,DE and DES were in the range of 1×10-8-1×10-4?1×10-7-1×10-4?2.5×10-8-1×10-4 mol/L,the correlation coefficient was high,the detection limit was 1.0×10-9?2.1×10-8?8.2×10-9 mol/L.The experimental data showed that the method was qualitative,quantitative and accurate,the sensitivity and detection limit were high,which provided a new idea for the detection of estrogen.Simultaneously,the actual sample detection using a new sample pretreatment method-aqueous two-phase extraction method.Using 70%volume fraction of ethanol as the extract,K2HPO4 was added to form a stable two-phase system.The actual sample detection did not require complex follow-up purification steps,and the method was simple and rapid,the biggest advantage was in the environment-friendly,non-toxic harmless.
