Chemiluminescence is widely applied in analytical chemistry owing to the unique extraordinary properties,such as no background interference and high sensitivity.Of which,the system composed of Bis?2,4,6-trichlorophenyl?oxalate and hydrogen peroxide?TCPO-H2O2?is not only suitable for detection of small molecular but also biological molecular due to unique characteristics,such as high quantum yield,few impurity disturbances and neutral pH.As one of nanometer materials,mesoporous silica nanoparticles?MSNs?are widely used in biological and analysis domain owing to some extraordinary properties,especially large mesopore volume,tunable porous channel,the surface easy to be modified and good biocompatibility.Graphene oxidation quantum dots?GOQDs?not only keep the original grapheme oxide?GO?biocompatibility,low toxicity and other excellent characteristics,also show a strong chemiluminescence enhancing effect.In this thesis,the work of development and application of chemiluminescent analytic method.The main contents include the following parts:?1?Based on the chemiluminescence resonance energy transfer system,we developed the strategy of triple signal amplification biosensor and it was characteried by the Fourier transform infrared spectroscope,UV-visible spectrophotometer,Zeta potential and so on.The detection results demonstate that a good linear relationship between the ratio of chemiluminescence intensity ?I and the logarithm of miRNA-21 concentration was obtained in the range of 0.00550 pmol L-1 in the pH7.4 PB buffer system,with a detection limit of 0.002 pmol L-1.The recovery rate in the real samples?human serum?was 93.6%–108.8%.?2?On the strength of the chemiluminescence resonance energy transfer system,we created the strategy of detecting 4-nitrophenol with high sensitivity and high selectivity,which employed chemiluminescence enhancing effect of GOQDs.The experimental results demonstate that a good linear relationship between the ratio of chemiluminescence intensity ?I and the concentration of 4-nitrophenol was obtained in the range of 1.0 to 300 pmol L-1,with a detection limit of 0.3 pmol L-1.The recovery rate in the real samples?tap water and Xiang River?was 93.4%–107.9%.?3?On the basis of the chemiluminescence resonance energy transfer system,we exploited the strategy of determining glutathione?GSH?with simpleness,speedness and high sensitivity,which conbine GO?quencher?with GSH?reductant?.Theexperimental results demonstate that a good linear relationship between the ratio of chemiluminescence intensity ?I and the concentration of GSH was obtained in the range of 0.1 to 200 nmol L-1,with a detection limit of 30 pmol L-1.The recovery rate in the real samples?human serum?was 93.0%–103.0%.
