This paper mainly covers the following aspects:1.The importance of enantioseparation of chiral drugs,the progress of chromatographic separation techniques,the recent development of chiral stationary phases and the composite CSP derived from protein and cyclodextrin(CD)-bonded silica were reviewed.2.The ?-cyclodextrin(?-CD)and hydroxypropyl ?-cyclodextrin(HP-?-CD)bonded chiral stationary phases were synthesized using 3-glycidoxyproryl-trimethoxysilane(KH-560)as coupling agent.The synthesis conditions of the ?-CD bonded chiral stationary phases were optimized in the small and bulk amounts.The synthesis process in the small amount showed larger bonded amount,higher enantioseparation ability and better reproducibility.3.The ?-cyclodextrin(?-CD)and hydroxypropyl ?-cyclodextrin(HP-?-CD)bonded chiral stationary phases were prepared for the enantioseparation of benzoin and its analogues.The effects of the composition of mobile phase on the retention and enantioseparation were evaluated.The enantioseparation of naproxen on these two CSPs were also investigated.Naproxen obtained preferable enantioseparation ability.4.By using the 2,4,6-trichloro-1,3,5-triazine as a cross-linker,two composite chiral stationary phases(CSPs)derived from bovine serum albumin(BSA)and ?-cyclodextrin(CD)-bonded silica or hydroxypropyl ?-cyclodextrin(HP-?-CD)bonded silica were prepared.The obtained CSPs were packed by slurrying method.Then the composite CSPs were applied to the enantioseparation of tryptophan,hydrobenzoin,benzoin,and mandelic acid.The influences of pH value of the eluent,organic modifier,and column temperature on the retention and enantioseparation were discussed.For the tryptophan and mandelic acid,the effects of eluent pH value on the retention and enantioseparation were great,but for the hydrobenzoin and benzoin,the effects were inapparent.Tryptophan and hydrobenzoin obtained excellent resolution on the composite CSPs.The chromatographic results were compared with that on ?-CD-bonded or BSA-immobilized CSP.Furthermore,chiral recognition mechanism was studied further with the LSS and SDM-R models.
