| In order to establish the scientific classification and evaluation methods of Gastrodia elata Blume,and strengthen quality control and comprehensive utilization of resources of medicinal Gastrodia elata,in this paper,the chemical compositions of Gastrodia elata were determined and analyzed using high performance liquid chromatography(HPLC)and liquid chromatography-mass spectrometry(HPLC-MS)combined with measurement techniques,quality of different Gastrodia tuber were comprehensively evaluated based on active ingredient content and peak area of Gastrodia tuber.The main contents and results are as follows:1.Extraction of active ingredient of Gastrodia tuber.Using reflux extraction and ultrasonic extraction crushing combination of the active ingredient Gastrodia elata sample extraction,liquid chromatography results show that the extraction methods is better,the higher the extract gastrodin content.2.Measurement and analysis of the biochemical components of Gastrodia tuber based on HPLC-MSHPLC mobile phase and optimum detection wavelength and chromatography conditions were optimized.Then,using the optimized parameters measured Gastrodia elata main chemical composition of the sample,the results showed that the measurement method for chromatographic fingerprinting get clear,less interference between the peaks,can effectively separate and detect active ingredient content Gastrodia,can in a short time in the detection process is completed.Gastrodia elata use HPLC-MS sample chemical composition were measured and analyzed and found Gastrodia elata contains more than 77 kinds of chemical substances,including seven kinds of chemical substances are abundant,the main active ingredient content as high as 34.36% gastrodin.Gastrodia elata 50 kinds of main chemical components and mass spectrometry analysis,and found more than 20 kinds of chemical substances have been identified,there are more than thirty kinds of chemical substances have not been identifiedand requires further study.3.Chromatography Classification and Quality Evaluation of Gastrodia elata BlumeUsing HPLC method was determined to optimize the biochemical fingerprints with nine kinds of Gastrodia gastrodin content;the use of analysis software to calculate the similarity coefficient within populations and between different populations biochemical fingerprints;using cluster analysis and similarity coefficient of Gastrodia elata systematically classified;results showed nine kinds of Gastrodia can be divided into three different groups.ClassⅠinclude Gastrodia elata standards,Shimen Hunan,Hubei Yichang,Dabie Mountain in Anhui,Guizhou generous,Lueyang Shaanxi,Tibet Changbai Mountains in Jilin Gastrodia elata police stupid day.Biochemical fingerprinting Gastrodia elata this category are: gastrodin isolated peaks are higher than any other peak separation of all components.The first category includes Yunnan Yiliang Ⅱ Gastrodia elata,Gastrodia elata fingerprinting of this category are:There are two tall peaks(38.1% and 25.2% respectively of the total peak area)in 2.4 min and7.0 min at peak but gastrodin short(about 5%).The first category includes Ⅲ Sichuan Liangshan Gastrodia elata.Fingerprinting of this category is between Gastrodia elata GroupⅠ and Ⅱ class,as large as its 2.4 min peak and the corresponding peak Ⅱ separate class Gastrodia,7.0 min peak separation than the corresponding peak Ⅱ class Gastrodia elata little.But its peak is much higher than gastrodin Ⅱ class Gastrodia elata corresponding peaks corresponding to peak near the class Ⅰ Gastrodia elata.According gastrodin content and peak area of Gastrodia elata quality comprehensive evaluation,performance Ⅰ type biochemical fingerprints Gastrodia elata contain higher gastrodin,is an excellent species,which comes from the quality of Gastrodia Yichang,Hubei and Hunan Shimen best,Yichang to the mountains between Hunan Shimen may be more suitable for the cultivation of Gastrodia elata. |
