Monascus pigments are secondary metabolites of monascus Fujian provinces is rich in red yeast rice,of which Gutian red yeast rice is the most prestigious,red yeast rice is a local specialty resources in Fujian province,its in-depth research,the full development and rational use,would have a significant impact on the rapid development of the province’s economy.However,monascus strains producing capacity of color is generally low,which is the main reason to restriction monascus pigment widespread use,therefore,many scholars are committed to screening high-yield strains of pigments,optimized culture conditions to increase pigment production,but studies about the mechanism is relatively less.In this paper,twenty plant monacus are isolated from Nanping,Sanming,Longyan,Gutian,after fermentation liquid measuring its intracellular and extracellular total color value to obtain a high-yield pigment strains a3,as the target strain.Identified by morphological identification and molecular biology to determine the target strain is Monascus purpureus.Optimization the liquid fermentation conditions of strain a3.Glycerol,peptone,temperature were taken at three levels,intracellular and extracellular total color value as evaluation index,carried out three factors and three levels of orthogonal experiment.Orthogonal experimental results show that three factors order which effect liquid fermentation to produce pigment for strain a3 is glycerol content>peptone content>temperature,Among them,the impact of the glycerol and peptone content for intracellular and extracellular color value reached significant levels,while the temperature of its impact was not significant.The best solution is a combination A2B2C2,namely,glycerol content 8%,peptone content 1.5%and temperature 32℃.The single factor results showed that,optimum fermentation time is 9d,speed is 180 r/min,inoculum size is 6%,liquid volume is 100mL for strain a3.Extracted whole cell protein of strain a3 under different carbon sources,use two-dimensional electrophoresis technology find their differences protein,identified the differences protein by biological mass spectrometry,Successful identification of 27 different protein primarily correlation with synthesis and amino acids biological,protein translation,redox enzymes,glycolysis enzymes,18 proteins were up-regulated expression,9 proteins were down-regulated expression.
